Aberrant transcriptional and post-transcriptional regulation of SPAG5, a YAP-TAZ-TEAD downstream effector, fuels breast cancer cell proliferation

Valeria Canu, Sara Donzelli, Andrea Sacconi, Federica Lo Sardo, Claudio Pulito, Noa Bossel, Anna Di Benedetto, Paola Muti, Claudio Botti, Eytan Domany, Silvio Bicciato, Sabrina Strano, Yosef Yarden, Giovanni Blandino

Summary: SPAG5 is a crucial driver of cell mitotic spindle, and its overexpression in breast cancer is associated with poor prognosis. It is a direct target of miR-10b-3p and is involved in an oncogenic feedback loop with YAP and TAZ.

YAP/TAZ and EZH2 synergize to impair tumor suppressor activity of TGFBR2 in non-small cell lung cancer

Federica Lo Sardo, Claudio Pulito, Andrea Sacconi, Etleva Korita, Marius Sudol, Sabrina Strano, Giovanni Blandino

Summary: YAP and TAZ play important roles in non-small cell lung cancer (NSCLC) by regulating the expression of miRNAs, including miR-25, 93, and 106b. They cooperate with EZH2 to transcriptionally repress TGFBR2 and other tumor suppressor genes, promoting lung tumorigenesis. These findings suggest YAP/TAZ and EZH2 as potential therapeutic targets for NSCLC treatment.

Lemaigre, Gabrielle Bergers, Leo A. van Grunsven, Georg Halder

Summary: YAP/TAZ are not essential for liver development and regeneration in hepatocytes, but they indirectly contribute to liver regeneration by maintaining bile duct integrity and ensuring immune cell recruitment and function.

The Conundrum of Giglio Island: Unraveling the dynamics of an apparent resistance to COVID-19 - A descriptive study

Antonio Bognanni, Armando Schiaffino, Fulvia Pimpinelli, Sara Donzelli, Ilaria Celesti, Sabrina Strano, Elena Solari, Giorgia Schiaffino, Gabriele Solari, Domenico Solari, Serena Delbue, Marta Rigoni, Giandomenico Nollo, Greta E. Muti, Giovanna E. U. Muti Schunemann, Holger J. Schunemann, Giovanni Blandino, Aldo Morrone, Paola Muti

Summary: Despite extensive exposure to COVID-19, residents of Giglio Island, Italy did not develop any symptoms of SARS-CoV-2. The study found only one asymptomatic infection, which was not confirmed by nucleic acid amplification tests. Further investigations will focus on exploring possible protective factors or cross-reactivity to SARS-CoV-2.

A Mouse Model of Cholangiocarcinoma Uncovers a Role for Tensin-4 in Tumor Progression

Summary: A mouse model combining Kras(G12D) expression in cholangiocytes with DDC diet-induced inflammation accurately mimics the development of iCCA, showing similar histological features to human IPNBs and activating signaling pathways seen in human iCCA. TNS4, stimulated by KRAS(G12D), plays a crucial role in promoting tumor progression in this model.

CircRNAs: role in human diseases and potential use as biomarkers

Lorena Verduci, Emilio Tarcitano, Sabrina Strano, Yosef Yarden, Giovanni Blandino

Summary: Circular RNAs (circRNAs) are a unique class of endogenous RNAs characterized by a covalent loop structure, high stability, and tissue specificity. Their main mode of action is sponging microRNAs, but they may also have the ability to bind specific proteins and encode short proteins. CircRNAs play various functional roles in human diseases, serving as potential biomarkers due to their stable nature and remarkable tissue specificity.

Insights into Intra-Tumoral Heterogeneity: Transcriptional Profiling of Chemoresistant MPM Cell Subpopulations Reveals Involvement of NFkB and DNA Repair Pathways and Contributes a Prognostic Signature

Mario Cioce, Andrea Sacconi, Harvey Pass, Claudia Canino, Sabrina Strano, Giovanni Blandino, Vito Michele Fazio

Summary: Chemoresistance in malignant pleural mesothelioma (MPM) can be predicted by the expression of ALDH1A3. By studying cell subpopulations, it was found that ALDH(bright) cells exhibit greater resistance to chemotherapy drugs, providing insights into the mechanisms of tumor resistance to therapy.

Arachidonic acid drives adaptive responses to chemotherapy-induced stress in malignant mesothelioma

Mario Cioce, Claudia Canino, Harvey Pass, Giovanni Blandino, Sabrina Strano, Vito Michele Fazio

Summary: Malignant pleural mesothelioma (MPM) is highly resistant to current treatment options and has a poor prognosis. Treatment-induced stress triggers an adaptive response involving complex rearrangement of the MPM secretome, mediated by the acquisition of a senescence-associated-secretory-phenotype (SASP). Arachidonic acid (AA) is identified as an early mediator of this adaptive response and is associated with chemoresistance in MPM.

Initiation of hepatic stellate cell activation extends into chronic liver disease

Vincent De Smet, Nathalie Eysackers, Vincent Merens, Mina Kazemzadeh Dastjerd, Georg Halder, Stefaan Verhulst, Inge Mannaerts, Leo A. van Grunsven

Summary: Recent research has shown that both the initiation and activation transcriptional programs of hepatic stellate cells are active in mice and humans with chronic liver disease. This suggests that molecules involved in both phases are equally important in finding druggable targets for chronic liver disease.

The inner nuclear membrane protein NEMP1 supports nuclear envelope openings and enucleation of erythroblasts

Didier Hodzic, Jun Wu, Karen Krchma, Andrea Jurisicova, Yonit Tsatskis, Yijie Liu, Peng Ji, Kyunghee Choi, Helen McNeill

Summary: This study reveals for the first time the crucial role of nuclear envelope protein NEMP1 in erythropoiesis. The researchers found that NEMP1 plays an important regulatory role in nuclear envelope openings and enucleation process of red blood cells. The results provide a valuable mouse model for studying defective erythropoiesis.

Hippo signaling instructs ectopic but not normal organ growth

W. Kowalczyk, L. Romanelli, M. Atkins, H. Hillen, C. Bravo Gonzalez-Blas, J. Jacobs, J. Xie, S. Soheily, E. Verboven, I. M. Moya, S. Verhulst, M. de Waegeneer, L. Sansores-Garcia, L. van Huffel, R. L. Johnson, L. A. van Grunsven, S. Aerts, G. Halder

Summary: The Hippo signaling pathway is not essential for normal organ growth, and the overgrowth phenotypes associated with its activation result from abnormal genetic programs. In addition, the transcriptional activity of Hippo pathway effectors is not correlated with cell proliferation. A functional screen in Drosophila identified several target genes of the Hippo pathway that are required for ectopic overgrowth but not normal growth.

By using a systems biology approach, the authors demonstrate the dual use of regulatory sequences in achieving context dependence for wound repair and tumor growth. This research is valuable for researchers in the fields of gene regulation, developmental biology, and cancer research.
