miR-214-mediated downregulation of RNF8 induces chromosomal instability in ovarian cancer cells

Defective DNA damage response (DDR) is frequently associated with carcinogenesis. Abrogation of DDR leads to chromosomal instability, a most common characteristic of tumors. However, the molecular mechanisms underlying regulation of DDR are still elusive. The ubiquitin ligase RNF8 mediates the ubiquitination of H2AX and recruits 53BP1 and BRCA1 to DNA damage sites which promotes DDR and inhibits chromosomal instability. Though RNF8 is a key player involved in DDR, regulation of its expression is still poorly understood. Here, we show that miR-214 could abrogate DDR by repressing RNF8 expression through direct binding to 3-untranslated region (3 UTR) of RNF8 mRNA in human ovarian cancer cells. Antagonizing miR-214 by expressing its inhibitors in A2780 cells significantly increased RNF8 expression and thus promoted DNA damage repair. Consistent with the role of miR-214 in regulating RNF8 expression, the impaired DNA repair induced by miR-214 overexpression can be rescued by overexpressing RNF8 mRNA lacking the 3 UTR. Together, our results indicate that down-regulation of RNF8 mediated by miR-214 impedes DNA damage response to induce chromosomal instability in ovarian cancers, which may facilitate the understanding of mechanisms underlying chromosomal instability.

Summary: Anaplasmosis caused by Anaplasma marginale is a frequently reported tick-borne disease in tropical and sub-tropical countries. This study examined the prevalence and phylogenetic origin of A. marginale in cattle blood samples from two livestock regions in Punjab Province, Pakistan. The results showed a higher prevalence of A. marginale in cattle from Dera Ghazi Khan compared to Lodhran, and all cattle breeds in both districts were equally susceptible to the infection.

Summary: Raising small ruminants is crucial for farmers' income in Pakistan. This study investigates the seasonal and molecular prevalence of T. gondii in different breeds of goats in Khanewal district, Punjab, Pakistan. The results show a relatively low prevalence of T. gondii, with the highest prevalence observed in summer. Factors such as male goats, neighboring farms with other dairy animals, and water supply from pools are associated with higher infection rates.

Comprehensive Analysis of Colorectal Cancer Immunity and Identification of Immune-Related Prognostic Targets

A prognostic model was constructed and validated, showing promising potential of CYSLTR2 as an important immune target for the prognosis of colorectal cancer.

Diversity and Genetic Structure of Theileria annulata in Pakistan and Other Endemic Sites

Summary: The study found that the genetic structure of Theileria annulata in Pakistan is similar to that in Oman, Tunisia, and Turkey, showing a high level of genetic diversity. The genotypes of the parasite in these countries are geographically sub-structured. The parasite populations in Oman, Indian Subcontinent (Pakistan), Africa (Tunisia), and Europe (Turkey) overlap, indicating some level of parasite gene flow between neighboring countries.

Distribution, prevalence and diversity of ticks (Acari: Ixodidae) infesting one-humped camels from Southwest Punjab, Pakistan

Summary: In this study, a total of 593 ticks were collected from one-humped camels in two tehsils of Layyah district, Punjab, Pakistan, in order to investigate their diversity, prevalence, and distribution. The results showed that camels located in Tehsil Choubara had a significantly higher tick infestation rate compared to camels from Tehsil Layyah. It was also observed that older camels were more prone to tick infestation. Hyalomma and Rhipicephalus were the two genera identified, with Hyalomma genus being the most prevalent.

Association of single nucleotide polymorphisms in heat stress protein 70 (HSP70) and 90 (HSP90) with the susceptibility of Pakistani sheep breeds to hemoparasitic infections

Summary: The study investigated the genotype and allelic frequency of SNP at 222 G > A in HSP70 and at ex6-7390T22G in the HSP90 gene in 204 sheep from Punjab. The tetra-primer amplification refractory mutation system-polymerase chain reaction (T-ARMS-PCR) revealed significant genotype frequency variation in the HSP70 gene, while the allelic frequency remained unaffected. The study suggests the genotypic screening of sheep before selection as breeders to improve the profitability and sustainability of animal production systems in Pakistan.

Association of genotypes at rs438228855 in bovine SLC35A3 receptor gene of Pakistani cattle with the susceptibility to develop complex vertebral malformation

Summary: The aim of this study was to report the genotype and allelic frequency at rs438228855 in SLC35A3 receptor gene and its association with complex vertebral malformation (CMV) in enrolled Pakistani cattle. The results showed non-significant variations in allelic and genotype frequency at rs438228855 among the three cattle breeds. The heterozygous (GT) genotype was most abundant, followed by the wild type (GG) genotype, and the mutant genotype (TT) was not observed. The study also found significant differences in various hematological parameters between the cattle breeds, but no association with the genotype at rs438228855 was observed.

Epidemiology and Phylogeny of Anaplasma ovis with a Note on Hematological and Biochemical Changes in Asymptomatic Goats Enrolled from Four Districts in Punjab, Pakistan

Summary: The present study aims to report the molecular prevalence of A. ovis in 1200 asymptomatic goats from four districts of Punjab, Pakistan. The results showed that 184 out of 1200 (15%) goat blood samples were infected with A. ovis. Risk factors associated with the prevalence of A. ovis and phylogeny of the bacterium were also documented. This study provides important insights for developing integrated control policies against A. ovis.

Sub Chronic Toxic Effects of Ethanolic Pongamia pinnata and Eucalyptus camaldulensis Leaf Extracts on Hematological and Biochemical Parameters of Blood in the Male Albino Mice

Summary: This study investigated the effects of ethanolic extracts of Pongamia pinnata (L.) Pierre leaves and Eucalyptus camaldulensis Dehnh leaves on hematological parameters and biochemical components of blood serum in male albino mice. The results showed that the P. pinnata leaf extract significantly increased cholesterol and triglyceride levels, while the E. camaldulensis leaf extract significantly reduced white blood cells, platelets, and alanine aminotransferase, and increased mean corpuscular hemoglobin levels. Both extracts showed a potential to disturb the blood biochemistry of healthy male albino mice, with more pronounced effects observed with the E. camaldulensis leaf extract.
