Summary: This study aimed to determine the pharmacokinetics of meloxicam in pigeons. The results showed that meloxicam was effectively and quickly absorbed after oral and intramuscular administration, with high bioavailability. After intravenous administration, the elimination was faster. Formulating a dosing regimen for pigeons presented a significant challenge due to the lack of a precise analgesic threshold and notable differences in analgesic thresholds among various animal species. Higher doses or shorter dosing intervals are recommended for treating pain in pigeons, but further research is required to verify these recommendations.

Summary: Ketamine has gained attention for its rapid and effective antidepressant response. However, the parenteral administration of ketamine may lead to significant undesirable side effects and the development of a similar drug without these side effects has been challenging. Oral ketamine, with its different pharmacological profile, appears to be a viable alternative with similar treatment effects, lower cost, and fewer medical resources needed.

Summary: This study evaluated the miscibility of cellulose derivatives to enhance the release rate and stability of microparticles containing a weakly basic drug, itraconazole (ITZ). The effect of organic acids on the microenvironmental pH (pHm) and ITZ release was also investigated. In a rabbit model, the synergistic effect of cellulose-based microparticles and pHm modulators on ITZ bioavailability was compared with the reference product. The results showed that ITZ, hydroxypropyl methylcellulose, and hydroxypropyl methylcellulose phthalate exhibited miscibility and the stability of microparticles was maintained for 6 months. Integration of cellulose-based microparticles and pHm modulators significantly increased the flux and relative bioavailability of ITZ and its active metabolite OH-ITZ.

Summary: Intranasal delivery is a popular non-invasive mode of administration due to its potential for targeted delivery to the brain. The anatomical connection of the nasal cavity with the central nervous system, as well as the high vasculature of the respiratory area, enables systemic absorption and avoids hepatic metabolism. Modeling the absorption events in the nasal cavity is challenging and requires sophisticated approaches.

Summary: In this study, a novel class of HIV-1 protease inhibitors with the free nitrogen or sulphone in morpholine cores as P2 ligands in combination with diverse substituted phenylsulfonamide P20 ligands was designed and biologically evaluated. These inhibitors exhibited prominent enzymatic inhibitory activity in low nanomolar ranges with relatively low cytotoxicity. In particular, inhibitor 1e demonstrated strong enzyme inhibitory activity, in vivo antiviral activity, and potency against drug-resistant variants. In silico studies provided molecular insights and predictions for further research.
