Pharmacokinetics and bioavailability of itraconazole oral solution in cats

Objectives The aim of this study was to describe the pharmacokinetics and bioavailability of itraconazole (ITR) oral solution in healthy cats. Methods The pharmacokinetics of ITR were studied in eight healthy, fasted cats after a single intravenous (IV) and oral (PO) administration at a dose of 5 mg/kg, in a two-period crossover design study. Blood was obtained at predetermined intervals for the determination of ITR concentrations with high-performance liquid chromatography. Pharmacokinetic characterisation was performed by a non-compartmental method using WinNonlin 5.2.1. Results After IV administration, the major pharmacokinetic parameters were as follows (mean SD): terminal elimination half-life (T-1/2z ) 15.8 +/- 1.88 h; area under the curve from time zero to infinity (AUC(0-)) 13.9 +/- 3.17 hg/ml; total body clearance 0.37 +/- 0.08 l/h/kg; apparent volume of distribution 8.51 +/- 1.92 l/kg; mean residence time 20.6 +/- 3.95 h. After PO administration, the principal pharmacokinetic parameters were as follows (mean +/- SD): T-1/2z 15.6 +/- 3.20 h; AUC(0-) 7.94 +/- 2.83 hg/ml; peak concentration 0.70 +/- 0.14 g/ml; time of peak 1.43 +/- 0.53 h. The absolute bioavailability of ITR oral solution after oral administration was 52.1 +/- 11.6%. Conclusions and relevance The disposition of ITR oral solution in cats is characterised by a long terminal half-life, a short peak time and moderate bioavailability.

Summary: A novel class of HIV-1 PR/RT dual inhibitors was designed and evaluated, showing significant improvement in HIV-1 RT inhibitory activity while maintaining comparable or improved potency against PR. Some inhibitors displayed excellent anti-HIV-1 activity, with a more reasonable ratio between PR and RT inhibition, providing new insight for further optimization.

Summary: Liver fibrosis is a common consequence of chronic liver diseases. Novel derivatives of 1-(substituted phenyl)-1,8-naphthalidine-3-carboxamide were synthesized, with compounds 17f and 19g showing inhibitory effects on fibrogenic markers and signaling pathways related to hepatic fibrogenesis. Compound 19g also demonstrated significant antifibrotic effects in vivo in BDL rats.

Summary: A sensitive method for determining nine herbicides in sediments and aquatic products was developed using QuEChERS and UHPLC-MS/MS, with satisfactory recoveries and linear calibration curves. While the residues of herbicides in paddy fish did not harm humans in the rice-fish co-culture system, there were ecological risks to fish, daphnia, and algae in the system.

Design, synthesis and biological evaluation of protease inhibitors containing morpholine cores with remarkable potency against both HIV-1 subtypes B and C

Summary: In this study, a novel class of HIV-1 protease inhibitors with the free nitrogen or sulphone in morpholine cores as P2 ligands in combination with diverse substituted phenylsulfonamide P20 ligands was designed and biologically evaluated. These inhibitors exhibited prominent enzymatic inhibitory activity in low nanomolar ranges with relatively low cytotoxicity. In particular, inhibitor 1e demonstrated strong enzyme inhibitory activity, in vivo antiviral activity, and potency against drug-resistant variants. In silico studies provided molecular insights and predictions for further research.
