While it is known that Retinoic Acid (RA) induces meiosis in mouse female fetal gonads, the mechanisms which regulate this process during spermatogenesis are poorly understood. We show that the All trans RA derivative (ATRA) and Kit Ligand (KL) increase meiotic entry of postnatal mouse spermatogonia in vitro without synergism. Competence to enter meiosis is reached by spermatogonia only at the stage in which they undergo Kit-dependent divisions. Besides increasing Kit expression in spermatogonia, ATRA also upregulates KL expression in Sertoli cells. Both ATRA and KL increase the expression of Stimulated by Retinoic Acid Gene 8 and Dmc1, an early meiotic marker. A specific Kit tyrosine kinase inhibitor prevents the increase in the number of meiotic cells induced by both the two factors, suggesting that they converge on common Kit-dependent signalling pathways. Meiotic entry induced by ATRA and KL is independent from their ability to affect germ cell viability, and is mediated by the activation of PI3K and MAPK pathways through Kit autophosphorylation. ATRA-induced phosphorylation of the two downstream kinases is mediated by a non-genomic mechanism. These data suggest that RA may control the timing of meiosis by influencing both the somatic and the germ cell compartment of the postnatal testis through the activation of the KL/Kit system.

Ultrasonic testicular size of 24,440 adult Vietnamese men and the correlation with age and hormonal profiles

Bac Nguyen Hoai, Long Hoang, Duc Tran, Thang Nguyen Cao, Luu Doan Tien, Andrea Sansone, Emmanuele A. Jannini

Summary: Investigating testicular size in men is crucial for understanding reproductive and sexual functions, but the lack of country-specific standard values hinders diagnosis and treatment. A retrospective study of 24,440 men in Vietnam revealed that hypogonadal and unexplained infertile men had significantly smaller testicular sizes compared to healthy fathers. Testicular volume in Vietnamese men was negatively correlated with LH and FSH levels, and positively correlated with testosterone levels.

Castration and emasculation in the Middle Age. The andrological conundrum of Peter Abelard

Andrea Sansone, Tommaso B. Jannini, Susanna Dolci, Emmanuele A. Jannini

Summary: Peter Abelard is a renowned figure in philosophy, theology, and psychology, known for his mastery of dialectical debate. His relationship with Heloise d'Argenteuil is characterized by intense passion and documented through their correspondence, which has become a model for sentimental education and romantic love letters. Through analysis of Abelard's writings and historical context, we aim to explore whether he was castrated or subjected to an even more brutal punishment.

PVA-Microbubbles as a Radioembolization Platform: Formulation and the In Vitro Proof of Concept

Valerio Da Ros, Letizia Oddo, Yosra Toumia, Eugenia Guida, Silvia Minosse, Lidia Strigari, Silvia Strolin, Giulia Paolani, Francesca Di Giuliano, Roberto Floris, Francesco Garaci, Susanna Dolci, Gaio Paradossi, Fabio Domenici

Summary: This proof-of-concept study establishes a delivery strategy for radioactive lanthanides against recurrent glioblastoma using biocompatible PVA microbubbles and endovascular radiopharmaceutical infusion. The successful formulation turns PVA microbubbles into a microdevice with efficient Yttrium cargo loading. The selective targeting of Yttrium-loaded microbubbles on glioblastoma-associated tumor endothelial cells is achieved through biorecognition between overexpressed alpha(V)beta(3) integrin and the ligand Cyclo(Arg-Gly-Asp-D-Phe-Lys) on the microbubble surface.
