Regulation of high mobility group box 1 and hypoxia in the migration of mesenchymal stem cells

Mesenchymal stem cells (MSCs) have been increasingly offered for tissue regeneration with the premise that they can survive and thrive amidst the microenvironment of injured or degenerate tissues. The role of high mobility group box 1 (HMGB1) and hypoxia in the proliferation and migration of rat bone marrow MSCs (rBM-MSCs) has been investigated. First, the effect of HMGB1 on the proliferation of rBM-MSCs was determined. Second, to evaluate the regulation of hypoxia and HMGB1 in the migration of rBM-MSCs, cells in the wound healing model were exposed to four conditions: normoxia (20% O-2) and complete medium, normoxia and HMGB1, hypoxia (1% O-2) and complete medium, hypoxia and HMGB1. RT-PCR and Western blotting were used to measure the expression of migration-related genes and proteins. HMGB1 inhibited the proliferation of rBM-MSCs; HMGB1 alone or together with hypoxia and promoted the migration of MSCs and upregulated the expression of HIF-1 alpha and SDF-1. These results demonstrated that HMGB1 arrested the proliferation of rBM-MSCs, but enhanced the migration of rBM-MSCs which could be further improved by hypoxia. This study strengthens current understanding of the interaction between MSCs and the microenvironment of damaged tissues.

Alloying and brushite coating improve corrosion resistance of magnesium in a simulated physiological environment

Summary: The study found that brushite coated JDBM had the highest corrosion resistance, followed by uncoated JDBM, and then pure Mg. The main corrosion product of the Mg alloys in the cell culture medium was MgCO3 center dot 3H(2)O. The corrosion form of the JDBM alloys was uniform corrosion, while that of pure Mg was not uniform.

Annular Defects Impair the Mechanical Stability of the Intervertebral Disc

Summary: This biomechanical study investigated the effects of cruciform and square incisions on the mechanical stability of bovine intervertebral discs. The results showed that both incisions reduced axial compressive and torsional stiffness, but did not affect flexion-extension and lateral-bending stiffness. This study is important for the development of new approaches to disc repair and patient rehabilitation after discectomy.

Summary: The use of three-dimensional (3D) printed biocomposite scaffolds, reinforced with magnesium oxide (MgO), has shown promising results in the treatment of critical-sized bone defects (CSD). The incorporation of magnesium ions (Mg2+) in the scaffold enhances mechanical performance and cytocompatibility, leading to improved bone regeneration and repair.

Identification of injury type using somatosensory and motor evoked potentials in a rat spinal cord injury model

Summary: This study investigated the electrophysiological and histopathological changes in contusion, distraction, and dislocation spinal cord injuries using a rat model. The results showed that different types of injuries caused different levels of damage to specific spinal cord regions, resulting in varying somatosensory and motor evoked potential responses. Additionally, the study found significant correlations between histological and electrophysiological findings and injury types. Intraoperative monitoring of somatosensory and motor evoked potentials has the potential to identify the type of iatrogenic spinal cord injury during surgery.

By delivering miR-146a-5p and miR-221-3p, the hUCMSC-EVs significantly suppressed LFH and inhibited the activity of the TGF-beta/SMAD4 signaling pathway.
