Our study shows that dendritic cells induce cytotoxic CD8(+) T-cell responses by cross-presenting tumor-derived peptides, and the proportion of membrane-derived neoantigens is associated with reduced survival and treatment response.

These chemokine clusters enhance T-APC interactions, prolonging the interaction time and optimizing controlled activation broadly throughout the tissue. Th1-intrinsic, interferon-gamma (IFN gamma)-dependent positive-feedback loop enhances the frequency and range of these clusters.

Summary: The contribution of XCR1+ dendritic cells (DCs) and SIRPa+ DCs in maintaining T cell function during exhaustion and immunotherapeutic interventions of chronic infections is poorly understood. In a mouse model of chronic LCMV infection, XCR1+ DCs were found to be more resistant to infection and highly activated compared to SIRPa+ DCs. Exploiting XCR1+ DCs through expansion or vaccination improved CD8+ T cell functionality and virus control. PD-L1 blockade was found to be more effective when combined with increased frequency of XCR1+ DCs, enhancing the functionality of exhausted CD8+ T cell subsets.

Summary: It has been found that donor MHC antigens are released from skin or cardiac allografts via extracellular vesicles (EVs) and captured by macrophages in lymph nodes or spleen, triggering B cell activation and DSA production. This unexpected role of graft-derived EVs opens up possibilities to interfere with EV biogenesis, trafficking, or function to restrict alloreactive B cell priming.

Summary: Conventional dendritic cells (cDCs) are a powerful type of antigen-presenting cells, consisting of different subsets with distinct developmental, phenotypic, and functional characteristics. After immunization, each subset of cDCs acquires antigens in the secondary lymphoid organs and presents them to CD4T cells in a spatiotemporal manner, resulting in multiple waves of antigen presentation. This review focuses on the kinetics of antigen presentation by each cDC subset and its impact on priming naive CD4T cells, as well as its implications in CD4T cell differentiation.

Summary: The study demonstrates the strong antitumor immune response of cisplatin-based nanoparticles in a tumor-bearing mouse model. The sustained retention of nanoparticles leads to persistent tumor MHC-I overexpression, promoting the formation of pMHC-I and enhancing the interaction with T cell receptors. Additionally, the nanoparticles upregulate OX40 on intratumoral CD8+ T cells and synergize with an agonistic OX40 antibody to suppress tumor growth by 89.2%.

Summary: By studying the spleens of nurse sharks, we found that sharks are capable of producing robust antigen-specific responses and affinity maturation of B cells, despite the absence of germinal centers (GCs) observed in other animals. We also identified immune-related marker gene expression in the spleen cells of sharks and tracked their localization in the splenic follicles. These findings provide insights into the evolutionary foundation of GCs.
