Summary: In this study, a new microcarrier with hyaluronic acid (HA) as the core material was developed for scalable serum-free suspension culture of hESC-derived MSCs. The optimal microcarriers, with a coating collagen concentration of 100 μg/mL or concave-structured surface (cHAMCs), allowed for the expansion of IMRCs within six days with a high yield. The harvested cells showed high viability, immunomodulatory and regenerative therapeutic promise, as well as increased secretion of extracellular matrix (ECM).

Summary: This study investigated the chondrogenic potentials of 3D high-density cultures of bone marrow and Wharton's Jelly-derived mesenchymal stromal cells (MSCs) supplemented with hTGF-beta 1. The study found that high-density cultures promoted chondrogenesis and that Wharton's Jelly-derived MSCs showed potential for chondrogenic commitment. These findings have important implications for cartilage regeneration and tissue engineering.

Summary: This study shows that the pluripotency and differentiation ability of human adipose-derived stem cells (hASCs) can be reversibly changed during sequential cultivation with 2D and 3D culturing processes, with at least a 2-fold enhancement in pluripotency observed in hASCs cultivated with a 3D culturing process after 2D culture. The environment of stem cell culture (2D or 3D cultivation) can greatly impact stem cell fate in terms of pluripotency and differentiation reversibility.

Summary: This study evaluated the effects of different size microplastics present in water bottles on human mesenchymal stromal cells. The results showed that microplastic treatments led to a decrease in cell proliferation, altered differentiation potential, and loss of osteocyte phenotype. This indicates that environmental microplastics can be bioavailable for cell uptake and may result in irreversible diseases.

Summary: This study investigated the impact of osteogenic differentiation on the immunomodulatory properties of hPDL-MSCs, and found that osteogenic differentiation reduced the ability of hPDL-MSCs to suppress CD4(+) T lymphocyte proliferation and enhanced this ability in the presence of IL-1 beta. Additionally, the expression and activity of indoleamine-2,3-dioxygenase and interleukin-8 were also affected by the differentiation process.

This work demonstrates the modulation of cell membrane potential and intracellular calcium signaling in undifferentiated stem cells upon optical excitation.

Summary: Wharton's jelly-derived mesenchymal stem cells (WJ-MSCs) possess multilineage differentiation potential and specific surface markers. In this study, RNA sequencing analysis revealed differentially expressed genes associated with apoptosis and identified upregulation of ZBTB16 and FOXO1, as well as downregulation of TGFA, in all differentiated groups. Additionally, several novel marker genes related to WJ-MSCs differentiation were identified. This study sheds light on the molecular mechanisms involved in long-term culture and differentiation of WJ-MSCs.

No significant changes were observed with inhibitors targeting different channels or signaling pathways, suggesting a unique mechanism for the vasorelaxant effect of alogliptin.

The guidelines were reviewed and approved by the Korean Academy of Medical Sciences.
