Summary: The ALDH2 rs671 mutation is a risk factor for the development and prognosis of atherosclerotic cardiovascular diseases. This study uncovers the important role of the ALDH2-Rac2 axis in efferocytosis during atherosclerosis, providing a potential direction for therapeutic strategies in cardiovascular diseases.

Summary: Pharmacological inhibition of TRPC3 channels by JW-65 represents a new antiseizure strategy with potential therapeutic benefits. JW-65 can impair the initiation of seizures, reduce susceptibility to epileptic seizures, and decrease sensitivity to antiepileptic drugs.

Summary: Atherosclerosis is the main pathological basis for cardio-cerebrovascular diseases, with abnormal accumulation of apoptotic and necrotic cells leading to plaque formation and rupture. The efficiency of phagocytosis of apoptotic cells in AS is low, and recent research has made progress in understanding the molecular mechanisms involved.

Summary: In atherosclerosis, macrophages play an important role in inflammation. CD68, a specific receptor in macrophages, regulates the occurrence and development of atherosclerosis. CD68 deficiency can reduce atherosclerosis, decrease inflammation and necrotic content, and increase smooth muscle cell content in atherosclerotic plaques.

Summary: This study investigates the role of TRPC3, a non-selective cation channel, in the regulation of insulin secretion and glucose control. The results show that TRPC3 is involved in glucose-stimulated insulin secretion and is functionally expressed in human and mouse islet beta cells. Activation of TRPC3 enhances insulin secretion and improves diabetes symptoms in animal models.

Summary: Clinical studies and structural analyses indicate a connection between higher saliva calcium levels and salivary stone formation. The mechanism of calcium overload during sialolithiasis is not well understood. This study shows that TRPC3 plays a crucial role in preventing the expression of calcification genes in salivary ductal cells, leading to elevated levels of calcium in saliva and the formation of salivary gland stones.

Summary: The research suggests that serum IRGM levels may be related to plaque rupture in STEMI patients, and deficiency of IRGM/Irgm1 can increase plaque stability by suppressing macrophage apoptosis through inhibiting ROS generation and MAPK signaling transduction. Targeting IRGM may represent a new treatment strategy for the prevention and treatment of acute cardiovascular deaths caused by plaque rupture.

Summary: This study explored the feasibility of using a home-made AV-nanobubbles (NBAV) for the detection of apoptotic macrophages and assessment of atherosclerosis lesions. In vitro experiments demonstrated that the NBAV had good stability and echogenicity. In vivo experiments showed that NBAV had strong and sustained echo enhancement in plaque areas, and NBAV-sensitive plaques exhibited more significant pathological changes and vulnerable plaque features.

Summary: The study reveals that TRPC3 is overexpressed in gastric cancer and correlates with malignant progression and poor prognosis. TRPC3 enhances Ca2+ entry and promotes NFATc2 nuclear translocation, contributing to cell proliferation and apoptosis regulation. TRPC3 may serve as a potential therapeutic target for gastric cancer intervention.

Summary: Increasing evidence suggests that hypothyroidism exacerbates atherosclerosis. Macrophage apoptosis is crucial in atherosclerotic plaque development. The study reveals that thyroid hormones attenuate oxLDL-induced macrophage apoptosis by activating the TR alpha 1-Erk1/2 pathway and inhibiting oxidative stress in a concentration-dependent manner, indicating a potential mechanism for hypothyroidism-accelerated atherosclerosis.

Summary: In patients with cardiovascular disease, there is a characteristic restriction in the intimal Ca2+ signaling pattern, indicating a distinct truncation of the inherent Ca2+ profile with CVD. This suggests that the unique Ca2+ signaling profile along the arterial endothelial network may play a role in the development of cardiovascular diseases.

Summary: The study identified a benzothiazole amide derivative as a TRPC6 activator and showed that compound 1s exhibited good anti-gastric cancer activity and inhibited invasion and migration of MKN-45 cells in vitro. The design and synthesis of similar compounds with potential TRPC6 inhibitory activity is discussed.
