Preparation and characterization of inclusion complexes formed between baicalein and cyclodextrins

This study investigates the solubility of baicalein (Ba) with the addition of modified cyclodextrins using phase solubility method. The solubility of Ba in the presence of natural (alpha-,beta-, and gamma-) cyclodextrins and its derivatives, namely, hydroxypropyl-beta-cyclodextrin (HP-beta-CD) and (2,6-di-O-methyl)-beta-cyclodextrin (DM-beta-CD), was higher than that of free Ba. In particular, the stability constant of inclusion complex with DM-beta-CD was 13672.671 mol(-1), which was the highest among the examined cyclodextrins. The inclusion complexes of Ba and DM-beta-CD were prepared via freeze-drying method, which were both characterized in the solution and solid state by UV-vis spectroscopy, differential scanning calorimetry (DSC), proton nuclear magnetic resonance (H-1 NMR), scanning electron microscopy (SEM), and X-ray powder diffractometry (XRPD). The UV-vis, DSC, H-1 NMR, SEM, and XRPD results proved the formation of inclusion complex between Ba and DM-beta-CD. Furthermore, the dissolution rate and thermal stability of the inclusion complex were significantly enhanced compared with the pure drug. Therefore, using DM-beta-CD can effectively improve the solubility and thermal stability of free Ba, which is a promising approach to promote its clinical application. (C) 2013 Elsevier Ltd. The method effectively removed interferences from samples and successfully quantified the target alkaloids using ion pair reverse phase high-performance liquid chromatography.

New Glycosyltransferases in Panax notoginseng Perfect Main Ginsenosides Biosynthetic Pathways

PnUGT94M1 regioselectively catalyzes the rhamnosylation of the C6 glucose of PPT-type ginsenosides Rg1 and Rh1, generating ginsenosides Re and Rg2, respectively, and is UDP-beta-L-rhamnose (UDP-Rha)-dependent.

Self-Double-Emulsifying Drug Delivery System Enteric-Coated Capsules: A Novel Approach to Improve Oral Bioavailability and Anti-inflammatory Activity of Panax notoginseng Saponins

Summary: In this study, self-double-emulsifying drug delivery system enteric-coated capsules (PNS-SDE-ECC) were used to enhance the oral bioavailability and anti-inflammatory effects of Panax notoginseng saponins (PNS). PNS-SDE-ECC significantly promoted the absorption of PNS in the intestinal tract and exhibited sustained release and stability. Compared to gastric capsules, PNS-SDE-ECC increased the relative bioavailability of NGR1, GRg1, GRe, GRb1, and GRd by 4.83, 10.78, 9.25, 3.58, and 4.63 times, respectively. Furthermore, PNS-SDE-ECC reduced OXZ-induced inflammatory damage in the colon by regulating cytokine expression.

The results showed that KXS improved learning and memory, prevented mitochondrial damage, alleviated myocardial injury, and reversed the expression of proteins involved in ferroptosis in the hippocampus and heart tissues.

Green synthesis of multifunctional carbon dots from Crataegi Fructus for pH sensing, cell imaging and hemostatic effects

Summary: This paper reports the synthesis of multifunctional carbon dots (CFCDs) using herbal medicine as a precursor, and investigates their fluorescent properties and biological activities. The obtained CFCDs have a graphitic structure, abundant functional groups, high aqueous solubility, and excellent stability. CFCDs exhibit pH-dependent fluorescence properties and low cytotoxicity, making them suitable for cell imaging. Moreover, CFCDs have good hemostatic effects and can promote platelet aggregation. CFCDs have potential for wide application in the medical domain.

A TLC-direct bioassay method for detection of anti-lipid peroxidation constituents from fruits of Perilla frutescens

Summary: A practical TLC-direct bioassay method was established for screening and evaluating antioxidant activity. The method involved a series of reactions, including the production of lipid peroxides by UV irradiation of linoleic acid and the conversion of peroxides into malondialdehyde. A dot blot test using TLC separation showed that substances with anti-lipid peroxidation activity appeared as white spots/zones on a red background. The method was validated for selectivity, sensitivity, linearity, precision, and recovery, and was successfully used to identify active antioxidant constituents from Perilla fruits.

Species difference in toxicokinetics and safety assessment of senecionine N-oxide in a UDP-glucuronosyltransferase 1A4 humanized mouse model

Summary: This study investigates the toxicity and toxicokinetic characteristics of a toxic PA N-oxide, senecionine N-oxide, using a UDP-glucuronosyltransferase 1A4 humanized mouse model (hUGT1A4 mouse model) and wild-type mice. Results show that senecionine N-oxide exposure caused less toxicity in hUGT1A4 mice, and N-glucuronidation occurred exclusively in hUGT1A4 mice. There were also differences in toxicokinetic characteristics between hUGT1A4 mice and wild-type mice, with enhanced clearance of senecionine N-oxide and senecionine in hUGT1A4 mice, leading to detoxification.

Medicinal and edible plant Allium macrostemon Bunge for the treatment of testosterone-induced androgenetic alopecia in mice

Summary: This study found that a plant called Allium macrostemon Bunge (AMB) promotes hair regrowth in AGA mice and identifies the underlying molecular mechanisms. AMB stimulates proliferation and migration of human hair dermal papilla cells (HDPC) and enhances growth factors expression through the activation of the Wnt/β-catenin signaling pathway. Treatment with AMB extract promotes hair regeneration in AGA mice. These findings contribute to the potential use of AMB in alopecia treatment.

High-throughput intact Glycopeptide quantification strategy with targeted-MS (HTiGQs-target) reveals site-specific IgG N-glycopeptides as biomarkers for hepatic disorder diagnosis and staging

Summary: Liver disease is a major cause of global mortality, and identifying biomarkers for diagnosing its progression is crucial for improving outcomes. Targeted mass spectrometry is a powerful tool for verifying biomarker candidates and clinical applications, particularly for glycoproteins translation. However, the limitation of analyzing only one sample per run has become apparent. In this study, a high-throughput intact N-glycopeptides quantification strategy was developed, allowing the validation of 20 samples per run with an average analysis time of 3 minutes per sample. The strategy was applied in a cohort of 461 serum samples and identified a panel of 10 IgG N-glycopeptides that have strong clinical utility in evaluating the severity of liver disease.
