Summary: Red ginseng and Ophiopogon japonicus are traditional Chinese medicines that have been used as food in China for thousands of years. These herbs were frequently used in traditional Chinese patent medicines, but their carbohydrate compositions were often disregarded during production, resulting in a significant amount of waste. By optimizing the extraction conditions, polysaccharides from Shenmai injection waste were obtained and further purified. The purified polysaccharides showed potential as prebiotics and antioxidants.

Summary: This study investigated the structure and antioxidant activity of Porphyra haitanensis polysaccharides (PHPs) extracted using different methods. Ultra-high pressure, ultrasonic, and microwave assisted treatments enhanced the total sugar, sulfate, and uronic acid contents of PHPs, particularly with ultra-high pressure treatment. These treatments also affected the monosaccharide ratio and physical characteristics of PHPs, resulting in a loose microstructure with increased porosity. All PHPs exhibited in vitro antioxidant capacity, with ultra-high pressure treated PHPs showing the strongest activity. Moreover, these PHPs effectively improved cell viability and reduced ROS levels in H2O2 induced cells, indicating their potential as natural antioxidants.

Summary: The purpose of this study was to improve the extraction of polysaccharide from Cercis chinensis Bunge leaves using ultrasound, and compare it with boiling extraction in terms of polysaccharide content, monosaccharide compounds, and bioactivity. The optimal conditions were determined to be ultrasound intensity of 180 W, extraction duration of 40 min, water to material ratio of 15:1 (g/g), resulting in a higher polysaccharide yield of 20.02 +/- 0.55 (mg/g) compared to boiling extraction (16.09 +/- 0.82 mg/g). The ultrasound-extracted polysaccharides exhibited higher antioxidant activity and contained more total sugars and uronic acids compared to the boiling method.

Summary: In this study, pectic polysaccharides (FDP and DDP) were obtained from fresh and dried Dendrobium officinale by extraction and purification. FDP had stronger scavenging capacity against radicals than DDP and both attenuated alcohol-induced liver injury in mice. FDP-treated mice showed lower transaminase levels, inflammatory cytokine expression, and higher antioxidant enzyme activities compared to DDP-treated mice. The results suggest that D. officinale pectin can protect the liver and fresh pectin with specific structural characteristics is more promising.

Summary: This study purified and characterized two polysaccharides (BCG-1 and BCG-2) from BCG-PSN, revealing that they are an α-D-(1 -> 4)-mannan with (1 -> 2)-linked branches and an α-D-(1 -> 4)-glucan with (1 -> 6)-linked branches, respectively. The bioactivity assays showed that BCG-1 and BCG-2 could dose-dependently increase the production of inflammatory mediators and have similar or stronger effects compared to BCG-PSN injection.

Summary: Liver disease is a major cause of global mortality, and identifying biomarkers for diagnosing its progression is crucial for improving outcomes. Targeted mass spectrometry is a powerful tool for verifying biomarker candidates and clinical applications, particularly for glycoproteins translation. However, the limitation of analyzing only one sample per run has become apparent. In this study, a high-throughput intact N-glycopeptides quantification strategy was developed, allowing the validation of 20 samples per run with an average analysis time of 3 minutes per sample. The strategy was applied in a cohort of 461 serum samples and identified a panel of 10 IgG N-glycopeptides that have strong clinical utility in evaluating the severity of liver disease.
