Orthotopic implantation mouse model and cDNA microarray analysis indicates several genes potentially involved in lymph node metastasis of colorectal cancer

In colorectal cancer (CRC) patients, metastasis to the regional lymph node (LN) is an important first step in the dissemination of cancers. To identify the genes possibly involved in LN metastasis of CRC, we analyzed LN metastases in an orthotopic implantation mouse model with 22 CRC cell lines using Matrigel, an extracellular matrix protein derived from mice sarcoma, and combined the data with gene expression profiles of cDNA microarray of those cell lines. With this implantation analysis, the incidence of LN metastasis was 60% in 228 orthotopically implanted mice and varied from 100% to 0% among the cell lines. KM12c and Clone A showed LN metastasis in all orthotopically implanted mice, but DLD-1, HCT-8, and SW948 did not show LN metastases at all. In contrast, the incidence of liver and lung metastasis in 22 CRC cell lines was 13% and 1%, respectively. Combining those data with cDNA microarray in vitro, we isolated 636 genes that were differentially expressed depending on the incidence of LN metastasis. Among those genes, the expression level of ring finger protein 125 (RNF125), previously known as an E3 ubiquitin ligase in T cell activation, was significantly different between primary tumors in Stage III CRC patients with LN metastasis and Stage II patients without LN metastasis. In conclusion, the orthotopic implantation mice model with Matrigel was useful, and we isolated candidate genes such as RNF125 that possibly play an important role in LN metastasis of CRC cells.

Protection of liver sinusoids by intravenous administration of human Muse cells in a rat extra-small partial liver transplantation model

Summary: Muse cells have a stronger ability to home to the liver and repair damaged tissue compared to MSCs, improving blood chemistry and proliferative hepatocytes within a short timeframe. Knockdown of hepatocyte growth factor and vascular endothelial growth factor-A can impair the protective effect of Muse cells on LSECs. Intravenous injection of Muse cells may be a feasible approach for LDLT with less recipient burden.

Child Nutrition in Disaster: A Scoping Review

Summary: Disasters pose a threat to the nutritional health of children, but adequate infant and young child feeding can prevent malnutrition and save lives. Despite progress in nutritional support during disasters, child malnutrition remains a concern. Factors such as malnutrition before disasters, food insecurity, living conditions in shelters, poor breastfeeding practices, sociocultural factors, and organizational challenges all impact child nutrition in disasters. It is recommended to adopt a coordinated approach that includes preparedness, policy development, and education for children, families, and relief workers. Periodic nutritional assessments and designated authorities are necessary for providing nutritional support. Education and involvement of the general population are also important. Future assessments should consider food allergies in children and the impact of nutrition on child mental health in disasters.

A study on hypothermia and associated countermeasures in tsunami disasters: A case study of Miyagi Prefecture during the 2011 great East Japan earthquake

Summary: This study analyzed the impact of hypothermia during tsunami disasters based on casualty data from Miyagi Prefecture during the Great East Japan Earthquake. The research examined the spatial distribution of hypothermia victims' locations and the effects of age group, location, environmental temperature, and tsunami exposure at different scales. It was found that hypothermia victims were only identified in coastal areas, indicating an increased risk of hypothermia due to wetting of the body caused by exposure to tsunami water and a cold environment at evacuation centers. The proportion of older individuals (>70 years) among hypothermia victims was significantly higher compared to drowning victims. Based on these findings, a response checklist in evacuation centers and a modified government response flowchart are proposed to reduce the risk of hypothermia after a disaster.

TWIST1 is a prognostic factor for neoadjuvant chemotherapy for patients with resectable pancreatic cancer: a preliminary study

Summary: TWIST1 is an important poor prognostic factor for pancreatic ductal adenocarcinoma (PDAC) patients who undergo adjuvant chemotherapy after radical resection. Positive expression of TWIST1 is associated with significantly poorer prognosis in terms of relapse-free survival and overall survival. This finding suggests that TWIST1 can be utilized as a predictive factor for the response to adjuvant chemotherapies in PDAC patients.

Infliximab Inhibits Colitis Associated Cancer in Model Mice by Downregulating Genes Associated with Mast Cells and Decreasing Their Accumulation

Summary: This study aimed to investigate the potential of anti TNF-alpha antibodies (Abs) in inducing colon cancer development and identify the genes involved in colitis-associated cancer. The results showed that TNF-a inhibited the proliferation, migration, and invasion of colon cancer cell lines, and the neutralization of TNF-a by anti TNF-a Abs was observed in vitro. In a mouse model of colitis-associated cancer, treatment with infliximab, an anti TNF-a Ab, significantly reduced the development of colon cancer. Furthermore, gene analysis revealed the role of mast cells in the development of cancer associated with colitis.
