Summary: This study demonstrated the importance of the Wnt/beta-catenin pathway in Chikungunya virus (CHIKV) infection. Using in vitro, in vivo, and ex vivo models, the study showed that CHIKV infection suppresses the active beta-catenin protein, leading to a reduction in cyclin-D1 and an upregulation of glycogen synthase kinase-3 beta. Inhibition of beta-catenin reduced viral RNA, protein, and new viral particles in vitro and reduced disease score and viral load in mice. Knockdown of beta-catenin also reduced viral particle formation. These findings provide mechanistic insights that may contribute to the development of future therapeutics for CHIKV infection.
