Harnessing the Intestinal Microbiome for Optimal Therapeutic Immunomodulation

Distinct cytotoxic agents currently used in the oncological armamentarium mediate their clinical benefit by influencing, directly or indirectly, the immune system in such a way that innate and adaptive immunity contributes to the tumoricidal activity. Now, we bring up evidence that both arms of anticancer immunity can be triggered through the intervention of the intestinal microbiota. Alkylating agents, such as cyclophosphamide, set up the stage for enhanced permeability of the small intestine, facilitating the translocation of selected arrays of Gram-positive bacteria against which the host mounts effector pTh17 cells and memory Th1 responses. In addition, gut commensals, through lipopolysaccharide and other bacterial components, switch the tumor microenvironment, in particular the redox equilibrium and the TNF production of intratumoral myeloid cells during therapies with platinum salts or intratumoral TLR9 agonists combined with systemic anti-IL10R Ab respectively. Consequently, antibiotics can compromise the efficacy of certain chemotherapeutic or immunomodulatory regimens. (C) 2014 AACR.

Harnessing the Vnn1 pantetheinase pathway boosts short chain fatty acids production and mucosal protection in colitis

Summary: This study shows that the overexpression of Vnn1 is associated with the severity of IBD. VIVA mice are resistant to experimental colitis, and the pantetheinase activity of Vnn1 has a protective effect on colonocytes, enhancing CoA regeneration and metabolic adaptation. It also promotes the production of microbiota-dependent short chain fatty acids, especially butyrate, which regulates mucosal energetics and is reduced in patients with IBD. The induction of Vnn1/VNN1 during colitis is a compensatory mechanism to strengthen the mucosal barrier, and enhancing vitamin B-5-driven metabolism may improve mucosal healing and increase the efficacy of anti-inflammatory therapy.

Leptospira interrogans Prevents Macrophage Cell Death and Pyroptotic IL-1b Release through Its Atypical Lipopolysaccharide

Summary: Leptospira interrogans is capable of infecting all vertebrates and causing leptospirosis, but they do not trigger cell death in macrophages and escape pyroptosis and apoptosis. Instead, they induce mild IL-1b secretion through nonlytic caspase 8-dependent gasdermin D pore formation. Pathogenic L. interrogans prevent caspase 11 dimerization and gasdermin D cleavage, thus escaping massive cell death. This novel mechanism allows leptospires to evade inflammation and potentially contribute to their stealthiness.

Microbiota-induced active translocation of peptidoglycan across the intestinal barrier dictates its within-host dissemination

Summary: Peptidoglycan, the major component of bacterial cell walls, plays a crucial role in affecting physiological and behavioral changes in mammalian hosts. However, the mechanism and regulation of its translocation from the gut to host tissues have not been fully understood. This study investigated the absorption and dissemination of peptidoglycan across the intestine. The results showed that peptidoglycan has a preference for certain host organs, particularly the brain, when absorbed through the gut. The translocation of peptidoglycan is an active process regulated by the microbiota-induced parasympathetic pathway through muscarinic acetylcholine receptors.

A neutrophil response linked to tumor control in immunotherapy

Summary: Neutrophils accumulate in solid tumors and their abundance is associated with poor prognosis. In this study, the role of neutrophils in immunotherapy and tumor control was investigated. The results demonstrate that successful therapies can expand tumor neutrophil numbers and thereby contribute to antitumor effects.

Summary: In this study, we found that adding losartan (LOS) to FOLFIRINOX (FFX) chemotherapy followed by chemoradiation (CRT) in patients with locally advanced pancreatic cancer (LAPC) resulted in a 61% R0 surgical resection rate. By evaluating the effects of neoadjuvant LOS on the tumor microenvironment, we identified potential mechanisms of benefit. The results showed that LOS+FFX+CRT reduced the expression of immunosuppression and pro-invasion genes and was associated with improved overall survival (OS).

Summary: The spatial distribution of tumor-infiltrating lymphocytes (TIL) can predict breast cancer outcome and response to treatment. A new method called ST-FFPE has been developed to analyze formalin-fixed paraffin-embedded samples, allowing for the study of archival tissue. By examining different tumor compartments, this method can characterize the cellular composition of the tumor microenvironment.

Summary: The intestinal microbiota plays a role in postnatal growth, and Lactiplantibacillus plantarum strain LpWJL has been found to support growth in undernourished animals. This strain stimulates insulin-like growth factor-1 (IGF-1) and insulin production and activity. Cell walls from LpWJL, as well as specific molecules, can promote growth despite undernutrition. NOD2 in intestinal epithelial cells is necessary for LpWJL-mediated IGF-1 production and growth promotion in malnourished animals.

Summary: In this study, a high-affinity immunomodulatory IM-T9P1-ASO was developed to target both TLR9 and PD-L1, enhancing the therapeutic efficacy of immune checkpoint inhibitors. The results showed that IM-T9P1-ASO can effectively inhibit tumor growth and promote anti-tumor responses, suggesting its potential importance for cancer treatment.

GTN Enhances Antitumor Effects of Doxorubicin in TNBC by Targeting the Immunosuppressive Activity of PMN-MDSC

Summary: Our study aimed to test the beneficial effect of the nitric oxide donor glyceryltrinitrate (GTN) in combination with the chemotherapy drug doxorubicin in the treatment of triple-negative breast cancer (TNBC). Using a mouse model of TNBC, we found that the combination of the two drugs induced greater tumor regression compared to monotherapies, which was correlated with stronger activation of the immune system and a decrease in immunosuppressive immune cells. These findings suggest a new therapeutic perspective for TNBC treatment.

Effects of a Lacticaseibacillus Mix on Behavioural, Biochemical, and Gut Microbial Outcomes of Male Mice following Chronic Restraint Stress

Summary: This study found that supplementation with a probiotic mix can alleviate anxiety responses in mice sensitive to stress and have certain effects on gut microbiota. The probiotic mix can attenuate the weight loss and coat abnormalities caused by chronic stress and reduce stress-related anxiety-like behaviors. Additionally, serum corticosterone levels are significantly higher in the probiotic group, and the expression of dopamine and serotonin transporters in the hippocampus is lower. There are few differences in bacterial genera proportions and cecal acetate levels between the groups.

Summary: This study reports data from a French case-control prospective multicenter study, which found differences in gut microbiota composition between NEC cases and controls. The relative abundance of Lactobacillus genus was significantly lower in the NEC group, while that of the Clostridium cluster III was significantly higher. The study also found a statistical association between gut microbiota and cesarean delivery and antifungal therapy.
