Summary: A study found that a new inhibitor XH30 demonstrates anti-tumor activity against TMZ-resistant GBM by inhibiting cell proliferation and inducing cell cycle arrest. XH30 also showed effectiveness in suppressing TMZ-resistant tumor growth in an animal model and enhancing TMZ cytotoxicity. Mechanistically, XH30 may exert its synergistic effect by repressing the key transcription factor GLI1 via the noncanonical hedgehog signaling pathway.

Summary: Liposarcoma is a common soft-tissue sarcoma with potential therapeutic targets such as MDM2 and CDK4. This study focuses on the response of liposarcoma to the MDM2 inhibitor nutlin-3, and identifies the involvement of proteostasis network nodes in the mechanism. The combination of nutlin-3 and proteasome inhibitors shows promising apoptosis induction, with the ATF4/CHOP stress response axis playing a crucial role. Activation of the unfolded protein response enhances the efficacy of nutlin-3. In vivo models further confirm the synergistic effects of idasanutlin and carfilzomib in liposarcoma growth.

This study found that the protein level of casein kinase 2a (CK2a) was increased in the endothelium of mice with type I diabetes (T1D). Inhibition of CK2a was found to accelerate skin wound healing in T1D mice by promoting endothelial cell proliferation via the Hedgehog signaling pathway.

Summary: This study aims to investigate the potential regulatory relationship between PTEN and Akt/mTOR inhibition. The results reveal that inhibition of mTOR suppresses PTEN expression by upregulating miRNAs, providing a novel explanation for the limited efficacy of mTOR inhibitors in the treatment of mTOR activation-related tumors. This study also suggests that dual inhibition of mTOR and miRNA is a promising therapeutic strategy to overcome the resistance of mTOR-related cancer treatment.

Summary: A study was conducted to investigate the inhibitory effect of polynitroxylated albumin (PNA) on leptomeningeal dissemination and metastatic tumor growth in a Sonic Hedgehog transgenic mouse model. The results showed that PNA treatment prolonged the lifespan of the mice and reduced the proliferation of primary tumors. Additionally, the number of cells in spinal cord metastatic tumors was significantly decreased in PNA-treated mice.
