Summary: This study explored the expression of Gal-9 and TIM-3 in AD patients and found that both Gal-9 and TIM-3 levels were higher in AD patients than in healthy controls. These levels were associated with disease activity, IgE levels, and circulating eosinophils and/or B cells. Furthermore, Gal-9 was found to inhibit T cell proliferation and induce apoptosis in AD patients.

Summary: The interaction between metalloproteins Mmp9 and Mmp14 and carbohydrate-binding protein galectin-3 plays a critical role in osteoclast-mediated bone resorption. The ability of Mmp9 and Mmp14 to degrade galectin-3 on the cell surface depends on its binding protein Lrp-1. These findings reveal a previously unrecognized galectin-3/Lrp1 axis that regulates both the transcriptional programs and intracellular signaling cascades essential for osteoclast function.

Summary: The regulation of protein O-GlcNAcylation plays a crucial role in stem cell pluripotency and differentiation. This study investigates the impact of O-GlcNAc modifications on extraembryonic endoderm (XEN) differentiation, focusing on the protein galectin-3. The results reveal that galectin-3 expression and secretion are influenced by O-GlcNAc modifications, but inhibiting global O-GlcNAcylation does not affect pluripotency or XEN differentiation in embryonic stem (ES) cells.

Summary: Galectin-3 plays a crucial role in the early pathogenesis of NAFLD by promoting hepatic steatosis through increased fatty acid uptake. Inhibition of Gal3 can reduce the occurrence of fatty liver, suggesting that targeting Gal3 may be a potential therapeutic approach for the treatment of NAFLD.

Summary: Metabolic deregulation in cancer cells promotes tumor growth and metastasis. In this study, PSPH, a key enzyme in serine metabolism pathway, was found to be upregulated in melanoma samples. Knocking down PSPH resulted in inhibition of melanoma growth and metastasis through suppression of 2-HG levels and subsequent activation of pro-oncogenic gene expression pathways.

Summary: Prostate cancer is the most common cancer among men worldwide, and hormone-related factors, such as androgens, estrogens, and their receptors, play a crucial role in its development and progression. Galectin-3, a family of proteins, is involved in various stages of tumor development and progression, but its expression regulation is still poorly understood. This review focuses on the interplay between Galectin-3 and estrogen receptor signaling pathways in prostate cancer cells and proposes a molecular mechanism involving ER, Galectin-3, and beta-catenin in the modulation of nuclear transcriptional events. Further understanding of the role of Galectin-3 and ER in prostate cancer could lead to the discovery of new drug targets to treat castration-resistant prostate cancer.

Summary: Spirulina polysaccharide (Sp) exhibits inhibitory effects on galectin-3 modulation and cell proliferation in gastric cancer cells. It also shows significant inhibition of hemeagglutination mediated by galectin-3 and provides cytoprotective effects. These findings suggest the potential therapeutic application of Sp in gastric cancer management.

Summary: Gal-3BP is identified as a highly secreted protein in PDAC and its overexpression is closely related to cell proliferation, migration, and invasion. It is found that Gal-3BP enhances galectin-3-mediated signaling, leading to increased PDAC metastasis. Blocking Gal-3BP with antibodies can effectively suppress PDAC metastasis.

Summary: Preventing or effectively treating metastatic uveal melanoma (UM) is critical due to its high occurrence rate and poor prognosis. The study identified a signaling pathway involving HGF, IGF-1, ARF6, ASAP1, and NFAT1 that promotes UM liver metastasis. Inhibiting this pathway could potentially be a targeted therapy for metastatic uveal melanoma.

Summary: This study investigates the mechanism of cigarette smoke-induced and galectin-3-mediated epithelial-mesenchymal transition (EMT) in airway epithelial cells (AECs). The results show that cigarette smoke extract (CSE) exposure affects the survival, morphology, migration, invasion, and clonogenicity of AECs, along with an increase in the expression of EMT markers, galectin-3, and RUNX-2. Inhibition or silencing of RUNX-2 downregulates galectin-3 and modulates EMT marker expression, migration, invasion, and clonogenicity in CSE-exposed AECs.

Summary: This study found that CD63 is highly expressed in vulvar squamous cell carcinoma (VSCC), and that CD9 is associated with tumor size, CD63 upregulation is associated with histological diagnosis and vascular invasion, while low expression of CD81 and CD82 is linked to worse prognosis. Furthermore, CD63 and CD82 may be potential therapeutic targets in VSCC.
