Summary: Gallbladder cancer (GBC) is a solid tumor with poor prognosis and current treatments are not effective. This study aimed to investigate a meaningful regulator in GBC. Zinc finger protein 64 (ZFP64) was identified as a vital regulatory protein in GBC. ZFP64 expression was higher in GBC tissues and correlated with poor prognosis. ZFP64 promoted migration, invasion, proliferation, anti-apoptosis, and epithelial mesenchymal transition (EMT) of GBC cells. Mechanistically, ZFP64 recruited HDAC1 to inhibit NUMB expression, leading to enhanced activation of the Notch1 signaling pathway. ZFP64 is expected to be a new focus for GBC prognostic marker and targeted therapy.

Summary: Gastric cancer stem cells (GCSCs) are important in the initiation and development of gastric cancer (GC). Circular RNAs (circRNAs) have diverse functions in cancer biology, acting as tumor suppressors or oncogenes. This study focused on the expression profile and functional roles of circRNAs in GCSCs. Through enrichment of GCSCs using a spheroid formation assay, it was found that circSLC4A7 was one of the most upregulated circRNAs in GCSCs. Further experiments showed that circSLC4A7 promoted CSC-like properties and cell proliferation, migration, and invasion by interacting with HSP90 and activating the Notch1 signaling pathway in GC.

Summary: This study analyzed the dysregulated profile of circular RNAs (circRNAs) in papillary thyroid cancer (PTC) tissues and found that circAGTPBP1 was upregulated and correlated with tumor characteristics. The downstream targets of circAGTPBP1 were identified as miR-34a-5p and NOTCH1. Functional experiments showed that circAGTPBP1 knockdown inhibited PTC cell migration, invasion, and metastasis, and miR-34a-5p inhibitor reversed these effects. CircAGTPBP1 knockdown also inhibited tumor growth in vivo, suggesting its potential as a therapeutic target for PTC.

Summary: Kindlin-2 plays critical roles in the development and homeostasis of multiple key organs, and our study reveals its importance in NOTCH-mediated physiological angiogenesis. We demonstrate that KINDLIN-2 enhances various functions of endothelial cells induced by VEGF-A and is crucial for maintaining the integrity of NOTCH1. The deficiency of KINDLIN-2 leads to impaired angiogenesis and avascular retinas, which can be rescued by the inhibitor DAPT, and we also find that KINDLIN-2 is involved in hyperactive angiogenesis stimulated by high glucose and could be a potential therapeutic target for diabetic retinopathy.

Summary: Long noncoding RNAs (lncRNAs) are noncoding RNAs with length greater than 200 nt, which have been extensively studied for their biological roles and mechanisms. Hypoxia is a proven microenvironmental factor that promotes solid tumor metastasis, with epithelial-mesenchymal transition (EMT) being one of the major mechanisms induced by hypoxia. Many lncRNAs induced by hypoxia have been shown to interact with protein/protein complex and chromatin/epigenetic factors, contributing to metastasis.

RP11-367G18.1 interacted with histone acetyltransferase p300 and regulated lysine 16 acetylation on histone 4 (H4K16Ac), thereby promoting ccRCC metastasis and growth. RP11-367G18.1 was associated with poor prognosis and may serve as a therapeutic target for ccRCC.
