Summary: Cyclins not only activate CDK complexes, but also serve as docking scaffolds for CDK substrates and inhibitors. G1-cyclins in yeast play a specific role in promoting bud growth and polarization, essential for cell survival. The discovery of a specific docking motif in G1-cyclins expands our understanding of cyclin specificity mechanisms.

Summary: CDKs, cyclins, and CKIs play important roles in the cell cycle regulatory machinery, and dysregulation of their expression or function can contribute to tumorigenesis. While designing CDK inhibitors to target tumor cells shows promise, the non-canonical functions of CDKs pose challenges in their application. This review aims to explore the biology of CDKs and their contribution to tumorigenesis, and discuss the pros and cons of CDK inhibition in the treatment of human cancers.

Summary: CDK11 downregulation in T1D acts as a protective mechanism against inflammation-induced apoptosis, and it may regulate Cyclin D3 signaling.
CDK11 hemideficiency protects against T1D progression and affects pancreatic islet sensitivity to cytokine-induced apoptosis.
CDK11/Cyclin D3 tandem could be a potential new intervention target in T1D.

Summary: Treatment with neoadjuvant BRAF/MEK-targeted therapy showed improved outcomes in female patients compared to male patients in melanoma. Preclinical studies revealed impaired tumour activity in male mice after treatment, with higher androgen receptor expression. Pharmacological inhibition of androgen receptor signalling improved treatment responses in male and female mice, while induction of androgen receptor signalling impaired responses in male and female patients.

Summary: This study reveals the correlation between estrogen response and prognosis in cholangiocarcinoma (CCA). CCA was classified into two clusters based on estrogen response, and COMT and HSD17B1 were identified as responsible factors for the differential estrogen response between the clusters. An estrogen response risk score (ESRS) model was constructed for prognostic prediction, and altered pathways, applicable drugs, and drug sensitivity were analyzed specific to the estrogen response.

Summary: The study revealed that high expression of ESRRA is associated with poor prognosis in gastric cancer, and silencing ESRRA can reduce cell viability, migration, and invasion capacities in GC. The downstream gene DSN1 and signaling pathway CDC25C-CDK1-Cyclin B1 were identified as important mechanisms through which ESRRA affects GC development.

Summary: Menopause is associated with visceral adiposity, hepatic steatosis, and increased risk for cardiovascular disease. This study found that ovariectomy-induced metabolic derangement in adult female mice, and hepatic adropin, regulated by estrogen, was negatively correlated with adverse post-menopausal metabolic phenotypes, which were partially reversed with adropin treatment.

Summary: Subgroup analyses suggest that females may derive less benefit from VEGF monoclonal antibody bevacizumab than males in treating metastatic NSCLC. Estrogen may promote resistance to VEGF-targeted therapies by enhancing pericyte coverage and myeloid recruitment. Blockade of the estrogen receptor pathway could potentially enhance the effects of antiangiogenic therapy.

Summary: This study demonstrates that bacteria can induce hemocyte proliferation in Chinese mitten crabs, and EsCsn5 plays a negative regulatory role in this process by controlling the degradation of EsCyclin E. Knockdown of EsCsn5 enhances hemocyte proliferation and reduces survival rate in crabs, leading to increased bacterial concentration in the hemolymph.

Summary: Kindlin-2 is found to be crucial for promoting AR signaling and breast cancer progression. It physically associates with AR and Src, forming a complex that facilitates Src-mediated AR Tyr-534 phosphorylation and signaling, leading to enhanced breast cancer cell proliferation and migration. Depletion of Kindlin-2 suppresses AR signaling, resulting in diminished breast cancer progression.

Summary: This study characterizes the alteration of the gut virome in azoxymethane-induced colorectal neoplasia in mice and identifies important viruses associated with tumor growth. The research shows that there is a significant shift in viral profile when tumors first appear, and certain viruses from the genera Brunovirus, Hpunavirus are positively associated with tumor growth, while viruses from Lubbockvirus show a negative correlation with tumor growth. These findings provide strong evidence of the potential role of bacteriophage in the development of colorectal neoplasia.
