Statins potentiate cytostatic/cytotoxic activity of sorafenib but not sunitinib against tumor cell lines in vitro

The aim of this study was to investigate the potential cytostatic/cytotoxic effects of HMG-CoA reductase inhibitors and two orphan drugs registered for the treatment of advanced renal cell carcinoma, i.e. sorafenib and sunitinib against several different tumor cell lines. Cytostatic/cytotoxic effects were measured using crystal violet or MTT reduction assays. Cell cycle regulation was investigated using flow cytometry and Western blotting. The combination of lovastatin and sorafenib (but not sunitinib) produced synergistic cytostatic/cytotoxic effects against all tested tumor cell lines. In this study, an impairment of the protein prenylation, especially geranylgeranylation, resulted predominantly in the potentiation of the cytostatic/cytotoxic activity of sorafenib, in cell cycle arrest in G1 phase, and, in poor induction of apoptosis. Moreover, due to the fact that it has been well documented that sorafenib compromises the heart function, we studied the interaction of lovastatin and sorafenib using rat cardiomyoblast line H9c2. The combination showed strong synergistic cardiotoxic effects. Statins and tyrosine kinase inhibitors were used at doses that are achievable clinically, which makes the combination promising for future studies, especially in urooncology, bearing in mind possible cardiotoxic effects. Neutrophil-to-lymphocyte ratio predicts nodal involvement in unfavourable, clinically nonmetastatic prostate cancer patients and overall survival in pN1 patients

Summary: The study aimed to determine the clinical utility of neutrophil-to-lymphocyte ratio (NLR) in predicting nodal involvement in prostate cancer patients treated with radical prostatectomy. The retrospective study included 205 patients and found that higher NLR was associated with lower incidence of nodal spread. Additionally, higher NLR in nodal-positive patients was correlated with shorter survival.

Summary: This paper aimed to investigate whether the selective cannabinoid CB1 receptor agonist ACEA could reverse the effects of corticosterone (CORT) on depressive and overactive bladder potential. Through experiments on 48 female rats, it was found that ACEA could restore CORT-induced bladder dysfunction and depressive behavior, as well as regulate urine and other biomarkers.

Summary: Chitotriosidase (CHIT1) is an enzyme that regulates macrophages and their role in asthma development was investigated. The expression of CHIT1 was examined in lung tissues of individuals with severe, uncontrolled, steroid-naive asthma, and a chitinase inhibitor called OATD-01 was tested in a mouse model of chronic asthma. The results showed that inhibiting CHIT1 had beneficial effects, reducing inflammation and airway remodeling in asthma.

Arginase Inhibition Mitigates Bortezomib-Exacerbated Cardiotoxicity in Multiple Myeloma

Summary: This study aims to investigate the improvement of cardiac toxicity caused by multiple myeloma and the therapeutic drug bortezomib through the enhancement of endothelial function. The results showed that bortezomib exacerbated the cardiac toxicity caused by multiple myeloma, while an arginase inhibitor partially prevented this effect. Therefore, arginase inhibitors may offer promise in the prevention/treatment of cardiac toxicity in multiple myeloma.

This study found that fractional CO2 laser treatment effectively improved these symptoms and increased the quality of life for women. The treatment restored the correct structure and composition of the vaginal epithelium, leading to positive results in both objective and subjective evaluations.

Summary: Through whole-exome sequencing, it was found that metastases in nonmetastatic ccRCC patients mainly originated from venous tumor thrombus (VTT) rather than primary tumors. Pathological grading of VTT outperformed other indicators in risk assessment. A new prognostic tool, TT-GPS score, incorporating VTT grading, showed better discriminatory ability in predicting the survival of nonmetastatic ccRCC patients with VTT compared to previously reported models.

Summary: This study compared the characteristics and 12-month clinical outcomes of patients with MINOCA treated before and during the COVID-19 pandemic. The rate of MINOCA was lower in 2020 and in-hospital stroke rates were higher during the pandemic. Further evaluation is needed to assess the association between worse clinical outcomes of MINOCA patients treated during the pandemic and the increased risk for thromboembolic complications of SARS-CoV-2 infection.
