Summary: The study revealed that CD40 signaling in Clec4f(+) Kupffer cells triggers anti-CD40 antibody-induced liver toxicity. However, controlled erythrophagocytosis and the linked anti-inflammatory signaling by the endogenous metabolite heme can be exploited to reprogram liver macrophages and prevent necroinflammatory liver disease caused by high-dose administration of anti-CD40 antibodies.

Summary: The study found that anti-CD40 antibody can increase the number of CD3+CD56+ effector cells in CIK cells by promoting the maturation and activation of dendritic cells. In addition, anti-CD40 antibody can also increase the expression of CTLA-4 in CIK cells. For the antitumor response of DC-CIK cells against renal cell carcinoma cells, anti-CD40 antibody is superior to anti-CTLA-4 antibody. The CTLA-4 inhibitor ipilimumab can significantly increase the proportion of CD3+CD56+ cells in DC-CIK cells.

Summary: Myeloid-derived suppressor cells (MDSC) are immature myeloid cells with suppressive activity in cancer. In a study on advanced melanoma patients undergoing anti-PD-1 immunotherapy, higher levels of monocytic MDSC (MoMDSC) were found in responders compared to non-responders before treatment. There were no significant changes in MDSC levels during therapy. This study suggests that MoMDSCs may be a useful tool for monitoring the immune status of melanoma patients.

Summary: Combinatorial immunotherapy strategies using engineered nanoformulations and toll-like receptor 9 agonist CpG ODN have shown great promise for cancer therapy. In this study, a self-assembled nanoparticle called CNPs was developed to deliver CpG ODN into dendritic cells, effectively stimulating immune response. Additionally, the CNPs-adjuvanted vaccine based on melanoma antigens and neoantigens exhibited both cellular and humoral immune responses, inhibiting tumor growth. CD16 CAR-T cells showed potential as a synergistic strategy for solid tumor immunotherapy.

Summary: Triple-negative breast cancer (TNBC) is a subtype of breast cancer with poor prognosis and limited targeted therapy options. Immunotherapies are emerging as novel treatments for TNBC, but they can also lead to the selection of resistant cancer cells and immune escape. Mice studies have shown that a vaccination strategy with dormant, immunogenic breast cancer cells and depletion of myeloid-derived suppressor cells (MDSCs) can elicit an effective immune response and induce tumor dormancy, providing a potential therapeutic opportunity.
