Summary: Deficiencies in DOCK8 result in quantitative and qualitative defects in IL-10-producing Breg cells, potentially due to abnormal Dock8(-/-) CD4(+) T cells. The study found that recombinant murine IL-21 can restore Bregs function and reduce inflammatory cell infiltration in the lung in a murine asthma model. This provides new insights into potential therapeutic strategies for allergic diseases, such as asthma associated with DOCK8 deficiency.

Summary: We investigated the effects of Treg cells on B. burgdorferi infection in BALB/c mice, which have a different immune response compared to C57BL/6 mice. Depletion of Treg cells before infection caused increased joint swelling at low infection doses, but did not significantly affect histopathology. Mice with higher borrelial load in their hearts and higher levels of serum interleukin-10 were observed when Treg cells were depleted before infection with a higher dose. Multiple animal models are necessary to understand the role of Treg cells in the host response to B. burgdorferi due to the variability in Lyme disease presentation in humans.

Summary: Regulatory B cells (Bregs) have a crucial role in maintaining human pregnancy. This study found that the cell percentages of CD24hiCD38hi Bregs and CD24hiCD27+ Bregs, which have the potential to produce IL-10, are increased in human decidua compared with the mid-luteal phase endometrium. The study also revealed positive correlations between Bregs and the expression of dendritic cells (DCs) and natural killer (NK) cells.

Summary: It has been established that T cells play a significant role in anti-tumor immune response, while the contribution of B cells has been insufficiently studied. In this research, the proportions and characteristics of B cells in tumor draining lymph nodes (TDLNs) and non-TDLNs from oral squamous cell carcinoma patients were analyzed. The results showed that TDLNs had a higher proportion of B cells, especially immunosuppressive regulatory B cells, compared to non-TDLNs. The presence of regulatory B cells in TDLNs may hinder the effectiveness of novel cancer immunotherapies in head and neck cancer.
