Summary: This study developed a non-invasive liquid biopsy method for early detection of gastric cancer by using circular RNAs (circRNAs) as biomarkers. The researchers identified a panel of 8 circRNAs that could reliably distinguish gastric cancer patients from non-disease controls, even in early-stage patients. The decreased expression of circRNAs in serum samples suggested their tumor specificity.

Summary: Ovarian cancer is the fifth leading cause of cancer-related mortality in women worldwide. Despite limitations in traditional treatment methods and biomarkers, circulating tumor DNA (ctDNA) shows promise as a biomarker for ovarian cancer, offering potential for early detection and personalized treatment strategies. However, technical and biological hurdles still need to be addressed before integrating ctDNA assays into clinical practice for cancer screening.

Summary: This study optimized a CLDN18.2 molecular beacon with a stem-loop hairpin structure, which can rapidly recognize CLDN18.2 RNA with high resolution and selectivity. The beacon has been successfully applied to circulating tumor cell detection, showing 100% concordance with tissue biopsy results.

Summary: This study found that the level of cf mtDNA in radon-induced lung cancer patients was significantly higher than other study participants, and there was a significant difference in cf mtDNA levels in the blood plasma of healthy volunteers exposed and not exposed to high doses of radon. Overall, healthy volunteers living in areas with high radon levels had higher mtDNA copy numbers than lung cancer patients who were not exposed to high doses of radon.

Summary: Circulating DNAs are fragmented DNA in blood plasma, including cell-free mitochondrial and nuclear DNA, and are altered in various pathological conditions. Mitochondrial DNA is associated with inflammation and elevated in cancer patients. It induces pro-inflammatory conditions and activates growth factors. It also triggers a positive feedback loop and activates pro-proliferating signaling in the tumor microenvironment. This review discusses the potential prognostic biomarkers and therapeutic candidates in different cancers, particularly prostate cancer.

Summary: Through genome-wide analysis and validation, we identified four CpG sites (cg04486886, cg06712559, cg13539460, and cg27541454) as DNA methylation markers in colorectal cancer tissues. The diagnostic prediction model based on these markers can effectively distinguish colorectal cancer from polyp patients, and the md-score can robustly detect colorectal cancer. The cg27541454 may be a promising candidate noninvasive biomarker for early diagnosis of colorectal cancer.

Summary: This study explores noninvasive multianalytical biomarkers and constructs integrative models for risk assessment and gastric cancer (GC) screening. Methylation, mutation, and H. pylori-specific antibody panels were established and validated, and integrated models incorporating these biomarkers outperformed traditional models for risk assessment and GC detection. This study provides new insights for a more precise GC screening strategy in the future.

Summary: Mitochondrial DNA mutations play a role in the development of cancer, leading to the use of circulating cell-free mtDNA as a potential tumor biomarker. A new approach for profiling ccf-mtDNA mutations was developed based on systematic optimization of sequencing methodology. The study also highlighted the importance of proper processing of plasma samples for accurate detection of tumor-specific mtDNA mutations in cancer patients.

Summary: Neutrophils play a crucial role in innate immune response by executing various effector functions. When activated, they undergo cell death processes to ensure the proper elimination of pathogens and damaged cells, contributing to timely resolution of inflammation. Neutrophils' cell death mechanisms include apoptosis, pyroptosis, necrosis, and others, ultimately maintaining homeostasis in the organism.
