Summary: GPVI is a receptor expressed on megakaryocytes and platelets that plays a role in both arterial and venous thrombus formation, as well as in the formation of platelet-leukocyte complexes. It is emerging as a potential therapeutic target for venous thrombosis, pulmonary thromboembolism, and cancer metastasis due to its involvement in venous thrombus formation and thromboinflammation. Circulating soluble GPVI is also being explored as a biomarker for thrombosis-related complications.

Summary: This study aims to improve the clinical applicability of circulating tumor DNA (ctDNA) detection in pancreatic ductal adenocarcinoma (PDAC) and investigate the impact of blood-draw site and time point on the detectability and prognostic role of KRAS mutations. The study found that KRAS detection in pre- and postoperative blood and tumor-draining portal venous blood was significantly associated with shorter recurrence-free survival and identified as independent prognostic markers. KRAS ctDNA detection was also an independent adverse prognostic marker in both curative and palliative PDAC patients.

Summary: The aim of this study is to clarify the prognostic influence of venous resection of the portal vein (PV) or superior mesenteric vein (SMV) on long-term outcomes in patients with pancreatic ductal adenocarcinoma (PDAC) of the head with suspected vascular invasion. PV-SMV resection was related to lower overall survival. However, on multivariate analysis, margin status was a more important prognostic factor than PV-SMV resection and true pathological invasion for survival.

Summary: The aim of this study is to determine the potential significant tumor microenvironment (TME) immune markers of long-term survival in pancreatic ductal adenocarcinoma (PDAC) patients. Immunohistochemical staining was performed to characterize the TME using tissue microarray. The results showed that high density of CD8+ lymphocytes and low density of FOXP3+ and iNOS+ tumor-associated macrophages (TAMs) are predictors of good prognosis in PDAC patients.

Summary: In the past, patients with pancreatic ductal adenocarcinoma often underwent immediate surgery for tumor resection. Now, more and more patients receive chemotherapy (radiotherapy) before surgery. It is known that pancreatic cancer patients have a higher risk of developing thrombosis. However, the incidence of thrombosis in patients with pancreatic cancer during neoadjuvant chemotherapy is not well-studied. Few studies have investigated VTE incidence in this population, and it is unclear if these patients should use perioperative thromboprophylaxis. This narrative review summarizes the current evidence.

Summary: Self-reported family history of melanoma in first-degree relatives had an overall sensitivity of 71%, specificity of 79%, PPV of 31%, and NPV of 95%, with decreased accuracy for second-degree relatives. A personal history of melanoma was the only factor significantly associated with accuracy in self-reported family history of melanoma. Age, sex, estimated nevus count, and number of prior personal melanomas were not significant predictors. Dermatologists should educate patients on the differences between melanomas, keratinocyte carcinomas, and pre-cancers. Confirming self-reported family history of melanoma may improve risk assessment for patients undergoing screening.

Summary: ObjectiveAPPEASE is a phase I study that evaluated the safety, dosing, and efficacy of the combination of pembrolizumab and rivoceranib in patients with metastatic malignancies. The study showed acceptable toxicities, a disease control rate of 75%, and tumor shrinkage in patients who had previously progressed on pembrolizumab alone. This combination treatment has potential for further investigation in larger patient populations.
