Summary: This study investigates the effects of non-thermal atmospheric pressure plasma on human adipose tissue-derived stem cells and explores the feasibility of using optical spectroscopy as a non-destructive method for cell analysis. The results show that exposed stem cells maintain their stemness and short exposure to plasma is relatively non-toxic. In addition, the plasma can accelerate the cell cycle and promote proliferation, and reflectance spectroscopy proves to be an efficient method for evaluating treatment response.

Summary: This study reports on the antibacterial and antioxidant activities of two tetrandrine derivatives against clinically important bacteria. The derivatives showed selective activity against Staphylococcus aureus and exhibited good antioxidant activity against ABTS(·+) radical. The mechanisms involved in antibacterial activity were investigated through studies on cell membrane integrity, reactive oxygen species, and binding to bacterial DNA plasmid.

Summary: This study presents a label-free impedance-based approach for cell viability assessment by characterizing multiple electrical cellular phenotypes in a high-throughput manner, introducing a concept called the complex opacity spectrum for improved discrimination of live and dead cells. The approach successfully discriminated between living and dead cells in different experimental scenarios and provides highly accurate and consistent cell viability analysis, validated by commercial fluorescence-based flow cytometry.

Summary: The properties of six commonly used, commercially available, fluorescent dyes were compared in staining right-handed B-DNA and left-handed Z-DNA. All showed different degree of fluorescence turn-on in the presence of B-DNA, but very little in the presence of Z-DNA. The optimal range of dye-DNA ratios of DNA was determined. While these dyes do not provide a turn-on type probe for Z-DNA, staining between B- and Z-DNA using dyes such as SYBR Green I was shown to be useful in tracking the kinetics of conformational changes between these two forms of DNA. Finally, SYBR Green I showed unique circular dichroism patterns in 4 M NaCl that change in the presence of double stranded DNA, both in the visible and UV range.

Summary: Neuronal death during cytotoxic edema is influenced by the opening of large conductance Pannexin-1 channels. Blocking these channels reduces and delays neuronal death. Reactive oxygen species (ROS) produced during neuronal swelling activates Panx1 currents, and scavenging ROS is neuroprotective. Microglial processes, attracted by Panx1 opening and ATP release, have a neuroprotective effect on swelling neurons.

However, there is currently no guidance on the selection of a fit-for-purpose cell viability detection method. Summary: To expedite the discovery of anticancer drugs, a series of fluoroquinolone analogs were synthesized through functionalization of moxifloxacin, ofloxacin, and ciprofloxacin. Among them, compound IIIf showed the highest potency in the NCI-60 Human Tumor Cell Line Screening Assay, while compound VIb exhibited significant effects in the ofloxacin derivatives group. The two compounds demonstrated potent growth inhibition and induced apoptosis in various cancer cell lines, making them promising lead molecules for the development of cytotoxic agents from quinolone scaffolds.

Summary: The study demonstrated that nanoemulsions synthesized using N. Sativa L. tincture have potential antioxidant and anticancer effects, showing significant toxicity against ovarian cancer cells while having lower toxicity against normal cells. The pro-apoptotic effect of the nanoemulsion was confirmed through staining methods, suggesting its potential as an anticancer agent in future studies.

Summary: This study evaluated the effects of a single administration of BrdU on cell proliferation and apoptotic events in the prenatal cerebellum. Results showed that BrdU at various doses reduced mitotic figures and proliferating cells, with the most severe effects seen at higher doses. High doses of BrdU also led to the activation of apoptotic events, as shown by TUNEL assay and caspase-3 immunohistochemistry.

Summary: This study investigated the anticancer, antioxidant, and antibacterial activities of Lepidium sativum L. seed acetone extract (LSSAExt) alone and in combination with biogenic silver nanoparticles (AgNPs). The results showed that LSSAExt and LSSAExt+AgNPs exhibited good antibacterial effects, were non-toxic to splenic cells, and induced apoptosis in cancer cells.

Summary: Opportunistic mycoses caused by Fusarium species, such as fungal keratitis, have significant clinical and epidemiological implications. The sensitivity of fungal cells to antifungal agents is influenced by biofilm formation, with biofilm cells showing 100-fold higher minimum inhibitory concentrations compared to planktonic cultures. Understanding the characteristics of fungal biofilms is crucial for optimizing antimicrobial therapy in the presence of resistant mechanisms in mycelial fungi.
