Summary: HPV E7 sensitizes cells to inhibition of aurora kinases, while HPV oncogenes sensitize cells to inhibition of phosphoinositide 3-kinases. Research has shown that treatment with Alisertib combined with radiotherapy or a PI3K inhibitor can improve tumor response, suggesting potential for further clinical development of these combinations.

Summary: The study identified a potential therapeutic strategy for GBM by targeting proteasome inhibition, which showed high sensitivity in GSCs due to their reliance on increased protein synthesis rate and loss of autophagy. Combinatorial inhibition of proteasome and autophagy resulted in enhanced cytotoxicity in PTEN-expressing GSCs, suggesting a synthetic lethal approach for this subset of GBM patients.

Summary: New strategies are urgently needed to improve the dismal median survivals of only ~15-20 months for glioblastoma (GBM) patients after the current standard of care treatment. This study demonstrates the potential importance of PLK1 inhibitors as an adjunct to GBM therapy, with novel mechanisms of action identified for volasertib.

Summary: Glioblastoma is a common and malignant brain tumor with a short survival period and low cure rate. Research on resistance mechanisms related to DNA damage repair pathways, glioma stem cells, blood-brain barrier, and systemic treatment, as well as the development of localized treatments, could improve survival rates and quality of life for patients.

Summary: A better understanding of the transcriptional evolution of IDH-wild-type glioblastoma is crucial for optimizing treatment. Through RNA sequencing, this study demonstrates that glioblastomas mainly evolve through microenvironmental reorganization rather than molecular evolution of tumor cells. The changes in tumor composition over time, including alterations in neuron and oligodendrocyte marker genes and an increase in tumor-associated macrophages, are confirmed by single-cell RNA-seq and immunohistochemistry.

Summary: The RNA binding protein and m6A reader HNRNPA2B1 is overexpressed in breast tumors compared to normal tissue. Overexpression of A2B1 leads to tamoxifen and fulvestrant resistance, while knockdown of A2B1 restores endocrine sensitivity. Targeting A2B1 could be a potential therapeutic strategy to overcome acquired endocrine resistance in breast cancer.

Summary: Aurora-A kinase shows differential expression in normal Mullerian epithelium, benign and borderline serous and mucinous ovarian epithelial neoplasms, and malignant serous ovarian tumors. Nuclear expression of unphosphorylated Aurora-A is present in normal and benign neoplastic epithelium, but lost in malignant serous neoplasms. Further studies on the clinical implications and role of nuclear Aurora-A expression loss in ovarian tumors are warranted.

Summary: This article reviews the clinicopathologic and genetic features of early ependymal tumor with MN1-BEND2 fusion (EET MN1-BEND2), classical astroblastomas, and recently described related pediatric CNS tumors. The discovery of genomic DNA methylation profiling as a powerful tool for CNS tumor classification is also briefly discussed. The article concludes that although genomic DNA profiling has facilitated the discovery of new CNS tumor entities, traditional methods are still necessary for accurate classification and diagnosis.
