Summary: Cholangiocarcinomas (CCAs) are a highly heterogeneous group of liver tumors with distinct prognostic and therapeutic traits. Specific treatment modalities are required for different subclasses, depending on anatomical localization and disease stage, to improve patient outcomes. Comprehensive molecular characterization has paved the way for precision oncological approaches in the treatment of CCAs.

TAp73 Inhibits EMT and Cell Migration in Pancreatic Cancer Cells through Promoting SMAD4 Expression and SMAD4-Dependent Inhibition of ERK Activation

Summary: Transcriptionally active p73 (TAp73) inhibits tumor progression in pancreatic ductal adenocarcinoma (PDAC) by promoting canonical TGF-beta/Smad signaling and preventing non-canonical ERK1/2-mediated TGF-beta signaling. TAp73 suppresses epithelial-mesenchymal transition and upregulates the expression of SMAD4, which mediates the inhibition of ERK activation and cell motility. Measuring the levels of TAp73 and/or SMAD4 could help predict the TGF-beta pathway preference in PDAC patients.

Changes in the Acetylcholinesterase Enzymatic Activity in Tumor Development and Progression

Summary: The cholinergic system, specifically acetylcholine receptors (AChR) and acetylcholine (ACh), has been found to play a role in cancer development. Acetylcholinesterase (AChE), which regulates AChR by hydrolyzing ACh, has been observed to have a relevant role in cancer. However, there is controversy regarding AChE's participation in cancer, as its enzymatic activity can either increase or decrease in different tumors. This review focuses on analyzing the involvement of AChE in cancer progression and proposes AChE as a central regulator in the initiation and progression of cancer via the cholinergic system. Modulating ACh levels with AChE can regulate AChRs differentially, leading to diverse cancer events.
