Summary: In this study, the researchers identified ubiquitin carboxyl terminal hydrolase L3 (UCHL3) as a deubiquitylase of Yes-associated protein (YAP) in Anaplastic thyroid carcinoma (ATC). UCHL3 stabilizes YAP through its deubiquitylation activity, and its depletion leads to decreased ATC progression, stemness, metastasis, and increased sensitivity to chemotherapy. The study also revealed that TEAD4, which binds to YAP and DNA, activates UCHL3 transcription by binding to its promoter.

Summary: Anaplastic thyroid cancer (ATC) is a rare and deadly malignancy with a poor prognosis and limited treatment options. However, research on its carcinogenesis, development, and therapy is hindered by low prevalence and difficulty in obtaining tissue samples. To address this, we created a genetically engineered ATC mouse model (mATC) that closely mimics the dynamics and immunological microenvironment of human ATC tumors. This model provides a valuable tool for studying the mechanisms and potential treatments for ATC.

Summary: The tumor immune microenvironment (TIME) in anaplastic thyroid carcinoma (ATC) is a complex and diverse ecosystem. This study reveals the dynamic changes of TIME in both papillary thyroid carcinoma (PTC) and ATC at the single-cell level. The findings demonstrate the presence of immunosuppressive factors and decreased immune cell infiltration in ATC, providing insights for targeted therapy to activate the anti-tumor immune response.

In vivo studies show that intratumoral injection of NIS mRNA formulated in optimized LPm NP significantly increases NIS expression in ATC tumor tissue, resulting in increased iodine 131 accumulation and significant inhibition of tumor growth.

Improving the management of this deadly disease involves the use of molecular tests for targeted therapies, fast-track dedicated care pathways, and prompt initiation of treatment. Adjuvant chemo-radiotherapy is recommended for localized disease after surgery, while chemotherapy or targeted therapy combined with decompressive cervical radiotherapy is recommended for locally advanced or metastatic disease.

Summary: This study analyzed the dysregulated profile of circular RNAs (circRNAs) in papillary thyroid cancer (PTC) tissues and found that circAGTPBP1 was upregulated and correlated with tumor characteristics. The downstream targets of circAGTPBP1 were identified as miR-34a-5p and NOTCH1. Functional experiments showed that circAGTPBP1 knockdown inhibited PTC cell migration, invasion, and metastasis, and miR-34a-5p inhibitor reversed these effects. CircAGTPBP1 knockdown also inhibited tumor growth in vivo, suggesting its potential as a therapeutic target for PTC.

Summary: This study developed and characterized 6 novel ATC PDX models with 4 matching cell lines, each with distinct genetic profiles. The PDX models showed remarkable resemblance to the original patient surgical specimens and had strong concordance with the original tumor in terms of genetic characteristics. These models can serve as excellent tools for preclinical therapeutic trials.
