Enhanced enrichment and detection of thermotolerant Campylobacter species from water using the Portable Microbe Enrichment Unit and real-time PCR

An enhanced enrichment using the Portable Microbe Enrichment Unit (PMEU) with the microaerobic bubbling of broths was applied for the detection of thermotolerant Campylobacter species from water. This PMEU enrichment was compared with the conventional static enrichment of the international standard ISO 17995:2005. In addition, Campylobacter detection after enrichment using a real-time PCR detection was compared with colony counts. The tests with stressed Campylobacter jejuni cells in drinking water indicated that the PMEU enrichment yielded a significantly higher number of Campylobacter cells in the Bolton broth compared with the conventional static incubation. Application of the real-time PCR technique shortened the Campylobacter detection time. This combination of method modifications can be used for Campylobacter detection from water and adds methodological repertoire for the rapid survey and management of waterborne outbreaks.

Climate Change Impacts on Microbiota in Beach Sand and Water: Looking Ahead

Summary: Beach sand and water are both important for human health and their microbiology has been studied for decades. The World Health Organization recently recommended including recreational beach sands in monitoring for enterococci and Fungi. Global climate change is affecting beach microbial contamination, and the changing world and human travel contribute to the spread of endemic allochthonous microbiota. In the future, alternative microbial standards will be necessary to ensure the health of beach users from both sand and water contaminants.

Water disinfection with geopolymer-bentonite composite foam containing silver nanoparticles

Summary: Geopolymer-bentonite composite foam with silver nanoparticles was prepared and applied for water disinfection in this study. The results showed that the foam had a promising inactivation efficiency against Escherichia coli and intestinal enterococci bacteria, but a poor inactivation efficiency against somatic coliphage viruses. These preliminary findings demonstrate the potential of geopolymer foam composites for point-of-use water disinfection.

Detection and quantification of SARS-CoV-2 RNA in wastewater influent in relation to reported COVID-19 incidence in Finland

Summary: Wastewater-based surveillance is a cost-effective method for monitoring the COVID-19 pandemic at a population level. This study in Finland monitored SARS-CoV-2 RNA in wastewater samples from multiple treatment plants over a 10-month period. The detection rate of SARS-CoV-2 RNA in wastewater was 79%, and the surveillance proved to be a reliable indicator for COVID-19 incidence, independent of clinical testing strategies or individual testing willingness.

Active surveillance of antibiotic-resistant bacteria using a comprehensive one-health approach can help reduce their emergence and spread, reduce associated economic impacts, and guide antimicrobial stewardship programs. Wastewater surveillance of antibiotic-resistant bacteria provides composite samples for monitoring the total population, making it a promising tool for monitoring these pathogens. However, the clinical utility, sensitivity, and uniformity of wastewater surveillance of antibiotic-resistant bacteria remains poorly understood, especially in relation to clinical evidence in communities.

The findings demonstrated the usefulness of supplementary methods and suggested their wider implementation to enhance groundwater quality monitoring.

Genomic comparison revealed three clusters of isolates from different sources, suggesting the hospital environment as a source of transmission. Summary: Antimicrobial resistance (AMR) poses a global threat to human and animal health, and monitoring resistant bacteria and genes is crucial. However, monitoring of AMR in the environment is lacking, hindering our understanding of how resistant microorganisms are disseminated and the human health risks. This lack of knowledge includes the "normal" background levels of environmental AMR, high-risk environments for transmission, antibiotic concentration effects, and detecting non-circulating resistance genes. We need to address these knowledge gaps before implementing large-scale AMR monitoring in the environment.
