Colis tin is a venerable antibiotic whose fortunes have been revived by its excellent activity, the diminishing output of novel clinically effective antibiotics and the increasing importance of MDR infection in burn surgery, both in the civilian and military arenas. This review synthesizes current evidence on the usage of colistin in burn surgery including the structure-activity relationship; dosing, pharmacokinetics/pharmacodynamic (PK/PD), analytic methods, resistance and current research efforts into the redevelopment of this antibiotic, to distil recommendations for future research and clinical efficacy. Phenotypic and Genotypic Adaptations in Pseudomonas aeruginosa Biofilms following Long-Term Exposure to an Alginate Oligomer Therapy

Juliette L. Oakley, Rebecca Weiser, Lydia C. Powell, Julian Forton, Eshwar Mahenthiralingam, Philip D. Rye, Katja E. Hill, David W. Thomas, Manon F. Pritchard

Summary: The study showed that long-term exposure to OligoG CF-5/20 did not lead to extensive mutational changes in P. aeruginosa, but may decrease the pathogenicity of bacterial biofilm and improve its susceptibility to other classes of antibiotics.

A New Look at the Purported Health Benefits of Commercial and Natural Clays

Alexander Incledion, Megan Boseley, Rachael L. Moses, Ryan Moseley, Katja E. Hill, David W. Thomas, Rachel A. Adams, Tim P. Jones, Kelly A. BeruBe

Summary: Clays believed to have medicinal properties since prehistoric times are still used today as complementary medicines. Recent studies suggest that clays with antibacterial characteristics could serve as alternatives to antibiotics in combating antibiotic resistance. However, research indicates that the ion concentration in clay leachates is insufficient to have antibacterial effects, and all clay leachates displayed toxicity towards THP-1 monocytes.

Treatment of cutaneous basal cell carcinoma with combined laser extirpation and methyl aminolevulinic acid: five-year success rates

E. A. Azzopardi, W. Abdelrahman, E. Azzopardi, B. O'Leary, J. Yarrow, N. Miles, C. Barbara, L. Camilleri, M. T. Clementoni, M. Murison

Summary: Basal cell carcinoma is the most common cancer, and excisional surgery is commonly used for treatment. However, this study shows that carbon dioxide laser extirpation assisted by immediate photodynamic therapy can achieve a high clearance rate with better functional and cosmetic outcomes at a lower cost compared to wide local excision. Further investigation through randomized controlled studies is needed.

Quantifying the effects of antibiotic treatment on the extracellular polymer network of antimicrobial resistant and sensitive biofilms using multiple particle tracking

Lydia C. Powell, Muthanna Abdulkarim, Joana Stokniene, Qiu E. Yang, Timothy R. Walsh, Katja E. Hill, Mark Gumbleton, David W. Thomas

Summary: The study utilized multiple particle tracking (MPT) to quantify the effects of antibiotic treatments on the physical and mechanical properties of antimicrobial resistant (AMR) bacterial biofilms. Results showed size-dependent and strain-related differences in diffusion coefficient values of biofilms, and dose-dependent treatment effects induced by polymyxin B (PMB) in E. coli biofilms. The ability of MPT to assess diffusion and mechanical effects of antibiotic therapies within the AMR biofilm matrix was highlighted, providing a valuable tool for pre-clinical screening of anti-biofilm therapies.

A physicochemical assessment of the thermal stability of dextrin-colistin conjugates

Emilie Chiron, Mathieu Varache, Joana Stokniene, David W. Thomas, Elaine L. Ferguson

Summary: The attachment of polysaccharide carriers increasingly improves the precision delivery and effectiveness of protein and peptide drugs. The stability of polysaccharide-based conjugates is crucial for their protective efficacy, and research on dextrin-colistin conjugates showed significant release of saccharides during prolonged storage at elevated temperatures, without affecting antimicrobial activity.

Alginate oligosaccharides enhance diffusion and activity of colistin in a mucin-rich environment

Joana Stokniene, Mathieu Varache, Philip D. Rye, Katja E. Hill, David W. Thomas, Elaine L. Ferguson

Summary: A model was developed to study the impaired diffusion of antibiotics across mucin barriers and showed that mucin binding leads to significant loss and reduced effectiveness of antibiotics. The use of mucolytics improves antibiotic diffusion and antimicrobial activity.

Summary: This study reports a non-antibiotic approach using naturally occurring and semisynthetic compounds to treat chronic skin wounds by modifying bacterial growth and inducing biofilm disruption. The compounds were found to stimulate immune responses and exhibit antimicrobial activity, leading to the resolution of infection and wound healing.

A Systematic Review and Meta-Analysis of the Clinical Outcomes for Adjunctive Physical, Chemical, and Biological Treatment of Dental Implants With Peri-Implantitis

Summary: This systematic review assessed the effectiveness of adjunctive therapies for treating peri-implantitis. Meta-analyses were performed on studies comparing conventional surgical or nonsurgical mechanical debridement with the addition of adjunctive therapies. The results showed that chemical therapy had significant effects in reducing probing pocket depth and increasing radiographic bone level, while no significant improvements were found in reducing bleeding on probing. The limited availability of standardized, controlled studies and heterogeneity between studies restrict the evidence for the benefits of adjunctive therapies.

Role of Enzymic Antioxidants in Mediating Oxidative Stress and Contrasting Wound Healing Capabilities in Oral Mucosal/Skin Fibroblasts and Tissues

Summary: Unlike skin, oral mucosal wounds heal rapidly and with minimal scarring, possibly due to the enhanced healing properties of oral mucosal fibroblasts (OMFs). This study compared oxidative stress biomarker and antioxidant profiles between oral mucosal/skin tissues and OMFs/skin fibroblasts (SFs) to investigate the role of antioxidant capabilities and oxidative stress in the preferential healing of oral mucosa.
