The species in Lobaria sect Lobaria are circumscribed based on secondary metabolites and the morphology of vegetative propagules such as isidia and soredia Lobaria pulmonaria typically forms soredia, which distinguish it from the other species of this section However, in L pulmonaria soredia may develop into isidioid soredia that can be confused with isidia In Macaronesia, this difficulty gave rise to controversial discussion about the taxonomic status of L pulmonaria var meridionalis, which forms cylindrical diaspores In the present study, a three loci approach (ITS, RPB2 and EF-1 alpha) was used to investigate the taxonomic status of L pulmonaria var meridionalis based on a phylogeny of Macaronesian taxa of the L pulmonaria group Phylogenetic analyses reveal that L pulmonaria v meridionalis is a distinct taxon, which is here proposed at the rank of species Lobaria macaronesica Cornejo & Scheidegger sp nov The new species is genetically distinct from the Asian L meridionalis, the cosmopolitan L pulmonaria and the Macaronesian L immixta

Long-read sequencing reveals the evolutionary drivers of intra-host diversity across natural RNA mycovirus infections

Deborah M. Leigh, Karla Peranic, Simone Prospero, Carolina Cornejo, Mirna Curkovic-Perica, Quirin Kupper, Lucija Nuskern, Daniel Rigling, Marin Jezic

Summary: In this study, PacBio long-read HiFi sequencing was used to characterize the intra-host populations of natural infections of the RNA mycovirus CHV1. The intra-host diversity of CHV1 was shaped by various factors including purifying selection, accumulation of mutations over time, and epistatic interactions within the host genome. Geographical landscape features impacted CHV1 inter-host relationships by restricting dispersal and causing founder effects.

A New Double-Stranded RNA Mycovirus in Cryphonectria naterciae Is Able to Cross the Species Barrier and Is Deleterious to a New Host

Carolina Cornejo, Sakae Hisano, Helena Braganca, Nobuhiro Suzuki, Daniel Rigling

Summary: A novel double-stranded RNA virus, CnFGV1, was characterized from the fungal species Cryphonectria naterciae, with 40% of tested strains being positive for the virus. The virus was able to transmit within-species and cross-species, inducing sectoring and growth reduction in infected strains. This study lays the groundwork for assessing the hypovirulence effect of CnFGV1 against the hornbeam decline in Europe.

Long-term preserved mycelium establishes the presence of Cryphonectria carpinicola in the Balkans and of Cryphonectria radicalis in Bulgaria

Carolina Cornejo, Mihajlo Risteski, Kiril Sotirovski, Daniel Rigling

Summary: In this study, we identified rare Cryphonectria-like species in our isolate collection from the Balkans using preserved dry mycelium and DNA data. We confirmed the presence of Cryphonectria carpinicola on a Carpinus tree for the first time in the Balkans, and extended the range of Cryphonectria radicalis on Castanea sativa to Bulgaria. The molecular analysis of the oldest specimen, dating back to 1998, confirmed the first reported finding of Cryphonectria radicalis in North Macedonia.

Revision of Dimelaena Norman (Caliciaceae, Ascomycota) species containing usnic acid reveals a new species and a new combination from China

Min AI, Qiuyi Zhong, Christoph Scheidegger, Lisong Wang, Xinyu Wang

Summary: Dimelaena tibetica is a new species characterized by a grayish yellow surface, usually with a covering of white pruina, a crustose thallus, areolate at center, radiate-plicate at margin, adnate apothecia and lacking gyrophoric acid. It differs from other usnic acid-containing Dimelaena species by its plane marginal lobes and adnate apothecia. Based on the study of fresh specimens from the environment of the type locality, Dimelaena altissima is proposed as a new combination. The phylogenetic position of the genus Dimelaena was confirmed by generating a sequence from a fresh specimen of Dimelaena oreina. All three species share common characters: usnic acid content and a yellowish green upper surface. Descriptions of these species were based on a combination of morphological and phylogenetic analyses. A key and phylogram are provided for these three Dimelaena species which produce usnic acid.
