Considering both the potential effects on human health and the need for knowledge of food composition, quantitative detection of synthetic dyes in foodstuffs and beverages is an important issue. For the first time, we report a fast quantitative analysis of the food and drink colorant azorubine (E 122) in different types of beverages using surface-enhanced Raman scattering (SERS) without any sample preparation. Seven commercially available sweet drinks (including two negative controls) with high levels of complexity (sugar/artificial sweetener, ethanol content, etc.) were tested. Highly uniform Au film over nanospheres (FON) substrates together with use of Raman signal from silicon support as internal intensity standard enabled us to quantitatively determine the concentration of azorubine in each drink. SERS spectral analysis provided sufficient sensitivity (0.5-500 mg L-1) and determined azorubine concentration closely correlated with those obtained by a standard HPLC technique. The analysis was direct without the need for any pretreatment of the drinks or Au surface. Our SERS approach is a simple and rapid (35 min) prescan method, which can be easily implemented for a field application and for preliminary testing of food samples.

Surface enhanced Raman spectroscopy-based evaluation of the membrane protein composition of the organohalide-respiring Sulfurospirillum multivorans

Dana Cialla-May, Jennifer Gadkari, Andreea Winterfeld, Uwe Huebner, Karina Weber, Gabriele Diekert, Torsten Schubert, Tobias Goris, Juergen Popp

Summary: The study focused on using surface enhanced Raman spectroscopy (SERS) to quickly identify the cofactors involved in the respiration process of S. multivorans. Through SERS analysis of membrane proteins from the bacterium under different cultivation conditions, it was found that S. multivorans cultivated with tetrachloroethene mainly contained Vitamin B-12, while those cultivated with other electron acceptors did not.

Towards translation of surface-enhanced Raman spectroscopy (SERS) to clinical practice: Progress and trends

Sandeep Surendra Panikar, Dana Cialla-May, Elder De la Rosa, Pedro Salas, Juergen Popp

Summary: Efforts have been made to develop stealth SERS substrates to maintain sensing capabilities in clinical samples by reducing background noise and enhancing sensing capability. Two important methods proposed are the use of zwitterionic molecules and nanoshearing force to achieve antifouling features for point-of-care-testing (POCT). Recent developments in SERS show reliable stealth and sensitivity for diagnosis or personalized treatment in complex clinical samples.

Aptasensor for the detection of Methicillin resistant Staphylococcus aureus on contaminated surfaces

Muhabat Adeola Raji, Ghadeer Suaifan, Atef Shibl, Karina Weber, Dana Cialla-May, Juergen Popp, Khaled Al-Kattan, Mohammed Zourob

Summary: There is growing evidence that contaminated hospital environments are crucial in transmitting nosocomial pathogens like MRSA. Infection control protocols rely on early laboratory detection of pathogens, but processing of environmental samples is less standardized than clinical samples. A novel pathogen aptasensor swab was developed to detect MRSA on non-absorbable surfaces, with a visual detection limit below 100 CFU/ml and a short 5-minute turnaround time. This selective tool shows promise in quick and cost-effective pathogen detection.

Summary: The timely diagnosis of MRSA is crucial for reducing morbidity/mortality, but current identification methods have shortcomings. A new optical immunosensor was developed to detect MRSA on contaminated surfaces, with high specificity and sensitivity, making it a powerful point-of-care diagnostic platform.

Coffee Ring Effect of Ag Colloidal Nanoparticles Dried on Glass: Impact to Surface-Enhanced Raman Scattering (SERS)

Petra Simakova, Eva Kocisova, Marek Prochazka

Summary: This study investigated the coffee ring formation of two types of Ag colloidal nanoparticles and their impact on SERS, revealing that h.-r. nanoparticles have highly enhancing sites for enormous SERS signal; despite the uneven particle and hot spots distribution, as well as SERS signal fluctuation, the method offers advantages of higher sensitivity measurement and spectral mapping over the dried pattern.

Cyclodextrin-assisted surface-enhanced Raman spectroscopy: a critical review

Natalia E. Markina, Dana Cialla-May, Alexey V. Markin

Summary: The use of cyclodextrins (CDs) in surface-enhanced Raman spectroscopy (SERS) is still developing with less than 100 papers published since 1993. The review reveals controversial information about the efficiency of CDs in SERS analysis, and further experimental investigations are needed to estimate their real potential in SERS-based analysis.

Aptamers: Potential Diagnostic and Therapeutic Agents for Blood Diseases

Maher M. Aljohani, Dana Cialla-May, Juergen Popp, Raja Chinnappan, Khaled Al-Kattan, Mohammed Zourob

Summary: Aptamers are RNA/DNA oligonucleotide molecules that specifically bind to targeted molecules. They have potential diagnostic and therapeutic applications for blood diseases and offer advantages over antibodies in terms of selection, production, modification, stability, and immunogenicity. Several aptamers have been developed and investigated in various biomedical areas. The pegaptanib sodium injection, an aptamer-based therapeutic, has been approved by the FDA for therapeutic use. Other aptamers are also in clinical trials. This review highlights the current status and promising aptamers in pre-clinical development for blood diseases.

TiO2/Ag nanostructured coatings as recyclable platforms for surface-enhanced Raman scattering detection

Adela Hankova, Anna Kuzminova, Jan Hanus, Petr Sezemsky, Radka Simerova, Vitezslav Stranak, Katarzyna Grochowska, Dujearic-Stephane Kouao, Katarzyna Siuzdak, Marek Prochazka, Tereza Kosutova, Ondrej Kylian

Summary: In this study, Ag/TiO2 coatings with different architectures were prepared using plasma-assisted techniques, and their SERS performance was investigated and compared. Nanoislands enhance the SERS signal, TiO2 degrades surface contaminants, and the photoactivity of titania enables the recycling of substrates. Nanostructured Ag/TiO2 coatings with larger active surface area show a good compromise between high detection limit and recyclability, allowing for the detection of methylene blue at low concentrations and easy cleaning by UV radiation.

Isidro Badillo-Ramirez, Jose M. Saniger, Jurgen Popp, Dana Cialla-May

Summary: Dopamine (DA) detection using SERS technique showed reproducible spectra in c-AgNP medium, while a completely different SERS profile was observed in h-AgNP solution. DA oxidation and conversion to PDA were found under different pH conditions, with in situ PDA formation triggered by laser irradiation. Long laser exposure time led to the formation of stable AgNP-PDA composites.
