Disulfiram targets cancer stem-like cells and reverses resistance and cross-resistance in acquired paclitaxel-resistant triple-negative breast cancer cells

Background: Triple-negative breast cancer (TNBC) has significantly worse prognosis. Acquired chemoresistance remains the major cause of therapeutic failure of TNBC. In clinic, the relapsed TNBC is commonly pan-resistant to various drugs with completely different resistant mechanisms. Investigation of the mechanisms and development of new drugs to target pan-chemoresistance will potentially improve the therapeutic outcomes of TNBC patients. Methods: In this study, 1-(4,5-Dimethylthiazol-2-yl)-3,5-diphenylformazan (MTT), combination index (CI)-isobologram, western blot, ALDEFLUOR analysis, clonogenic assay and immunocytochemistry were used. Results: The chemoresistant MDA-MB-231PAC10 cells are highly cross-resistant to paclitaxel (PAC), cisplatin (CDDP), docetaxel and doxorubicin. The MDA-MB-231(PAC10) cells are quiescent with significantly longer doubling time (64.9 vs 31.7 h). This may be caused by high expression of p21(Waf1). The MDA-MB-231PAC10 cells express high aldehyde dehydrogenase (ALDH) activity and a panel of embryonic stem cell-related proteins, for example, Oct4, Sox2, Nanog and nuclealisation of HIF2 alpha and NF-kappa Bp65. We have previously reported that disulfiram (DS), an antialcoholism drug, targets cancer stem cells (CSCs) and enhances cytotoxicity of anticancer drugs. Disulfiram abolished CSC characters and completely reversed PAC and CDDP resistance in MDA-MB-231PAC10 cells. Conclusion: Cancer stem cells may be responsible for acquired pan-chemoresistance. As a drug used in clinic, DS may be repurposed as a CSC inhibitor to reverse the acquired pan-chemoresistance.

LncRNA Tug1 maintains blood-testis barrier integrity by modulating Ccl2 expression in high-fat diet mice

Summary: Sertoli cells play a crucial role in spermatogenesis by forming the blood-testis barrier (BTB) and creating a unique microenvironment. This study reveals that a high-fat diet impairs BTB integrity and decreases sperm quality. The long noncoding RNA (lncRNA) Tug1 is found to be involved in the disruption of BTB by regulating Ccl2 and tight junction (TJ)-related proteins. Mechanistically, Tug1 directly binds to EZH2 and regulates H3K27me3 occupancy in the Ccl2 promoter region.

Osteopontin and Cancer: Insights into Its Role in Drug Resistance

Osteopontin (OPN) is a protein that is overexpressed in various types of tumors and is involved in multiple cellular processes related to drug resistance. Prohibitin Links Cell Cycle, Motility and Invasion in Prostate Cancer Cells

Summary: Prohibitin (PHB) is a tumor suppressor gene that has multiple molecular activities. Overexpression of PHB leads to cell cycle arrest in the G1/S phase and represses the androgen receptor (AR) in prostate cancer cells. PHB interacts and represses members of the E2F family, which may also be linked to AR, making the AR:PHB:E2F interaction axis highly complex. Knockdown of PHB using siRNA increased the growth and metastatic potential of LNCaP mouse xenografts. On the other hand, ectopic overexpression of PHB affected the expression of several hundred genes in LNCaP cells, including downregulated expression of WNT family members and cell adhesion pathways. Online data analysis showed decreased PHB expression in clinical cases of metastatic prostate cancer, and its expression was correlated with higher WNT expression in metastasis. Overexpression of PHB reduced cell migration, motility, invasion, and attachment in prostate cancer cells. Mechanistically, AR was found to be involved in the regulation of the WNT genes, and these WNTs were also regulated by the cell cycle. Therefore, the repressive effects of PHB on AR, E2F, and WNT expression suggest its importance in inhibiting prostate cancer metastasis.

nWASP Inhibition Increases Wound Healing via TrKb/PLCγ Signalling

Summary: This study found that nWASP expression was significantly increased in non-healing chronic wounds and its inhibition improved cellular behavior and wound healing. TrkB and PLC gamma 1 signaling pathways were identified as important regulators of nWASP. Therefore, nWASP is considered a new therapeutic target for non-healing wounds.

Vascular endothelial growth factor B regulates insulin secretion in β cells of type 2 diabetes mellitus mice via PLCγ and the IP3R-evoked Ca2+/CaMK2 signaling pathway

Summary: This study investigated the effect of vascular endothelial growth factor B (VEGFB) on regulating insulin secretion in type 2 diabetes mellitus (T2DM) and its underlying mechanism. The results showed that knockout of VEGFB exacerbated the decrease in insulin secretion in T2DM mice, while overexpression of VEGFB improved glucose tolerance and insulin sensitivity. VEGFB activated the Ca2+/CaMK2 pathway via VEGFR1-PLC gamma and IP3R pathway to regulate insulin secretion in T2DM.

The fiber structure evolution and performance changes of SiC-ZrC fibers after 1600 °C heat treatment

Summary: This study investigates the influence of different heat treatment atmospheres on the microstructural and mechanical behaviors of SiC-ZrC fibers. Experimental results show that the fibers' mechanical performance declines under vacuum and argon environments. Under vacuum conditions, the amorphous phase decomposes and crystallinity increases, but no clustering of ZrC phase is observed. In contrast, under argon conditions, the amorphous phase decomposes and clustering of ZrC particles occurs, resulting in inferior mechanical properties of the fibers.
