Quercetin downregulates Mcl-1 by acting on mRNA stability and protein degradation

BACKGROUND: We recently demonstrated that quercetin, a flavonoid naturally present in food and beverages belonging to the large class of phytochemicals, was able to sensitise leukaemic cells isolated from patients with chronic lymphocytic leukaemia (CLL) when associated with recombinant tumour necrosis factor-related apoptosis-inducing ligand (TRAIL) or anti-CD95. We also showed that quercetin potentiated the effect of fludarabine on resistant B cells from CLL patients. Resistance to therapy in CLL depends on the expression and activity of anti-apoptotic proteins of the Bcl-2 family. Among these, myeloid cell leukaemia-1 (Mcl-1) has been associated with apoptotic resistance in CLL. Therefore, we investigate here whether the sensitising activity of this flavonoid, which leads to increased apoptosis in both cell lines and CLL, could be related to Mcl-1 expression and stability. RESULTS: B cells isolated from CLL patients showed different levels of Mcl-1 protein expression, resulting, in several cases, in increased sensitivity to fludarabine. Quercetin significantly enhanced the downregulation of Mcl-1 in B cells isolated from selected patients expressing detectable levels of Mcl-1. In U-937 cells, quercetin increased Mcl-1 mRNA instability in the presence of actinomycin D. When cells were treated with MG-132, a proteasome inhibitor, Mcl-1 protein level increased. However, quercetin, in the presence of Z-Vad-FMK, continued to lower Mcl-1 protein expression, indicating its independence from caspase-mediated degradation. In contrast, co-treatment of quercetin and MG-132 did not revert the effect of MG-132 mono-treatment, thus suggesting a possible interference of quercetin in regulating the proteasome-dependent degradation of Mcl-1. Gossypol, a small-molecule inhibitor of Bcl-2 family members, mimics the activity of quercetin by lowering Mcl-1 expression and sensitising U-937 cells to apoptosis induced by recombinant TRAIL and the Fas-ligand. CONCLUSION: This study demonstrates that in U-937 cells, quercetin downregulates Mcl-1 acting directly or indirectly on its mRNA stability and protein degradation, suggesting that the same mechanism may bypass resistance to apoptosis in leukaemic cells isolated from CLL patients and sensitise B cells to apoptosis induced by drugs and death receptor inducers. A critical evaluation of risk to reward ratio of quercetin supplementation for COVID-19 and associated comorbid conditions

Summary: The interim results of large trials on COVID-19 using antiviral drugs show little to no effect, leading to a continued search for alternate drugs and supplements. This review highlights the possible efficacy of quercetin in treating and preventing severe acute respiratory syndrome coronavirus and its comorbidities. Multiple clinical trials are currently assessing the efficacy of quercetin and other drugs/supplements for COVID-19. Despite its therapeutic properties, further trials are needed to establish its clinical efficacy.

Phenolic Extract from Extra Virgin Olive Oil Induces Different Anti-Proliferative Pathways in Human Bladder Cancer Cell Lines

Summary: Regular consumption of olive oil is associated with protection against chronic-degenerative diseases, such as cancer. Epidemiological evidence indicates an inverse association between olive oil intake and bladder cancer risk. We investigated the anti-proliferative effects of a phenolic extract prepared from an extra virgin olive oil (EVOOE) on two human bladder cancer cell lines, and the results suggest that the mixture of phenolic compounds in extra virgin olive oil activates different anti-proliferative pathways.

Senolytic Flavonoids Enhance Type-I and Type-II Cell Death in Human Radioresistant Colon Cancer Cells through AMPK/MAPK Pathway

Summary: The role of autophagy and senescence in cancer resistance to ionizing radiation is still not fully understood. This study found that the flavonoids quercetin and fisetin can induce autophagy and senescence-associated inflammatory markers by activating AMPK and inhibiting the ERK pathway. The combined treatment promotes autophagy and senescence induction, lowers the threshold for cell death, and induces apoptosis.

Stillage Waste from Strawberry Spirit Production as a Source of Bioactive Compounds with Antioxidant and Antiproliferative Potential

Summary: The extract obtained from the waste of fermented strawberry distillate production exhibited high antioxidant and antiproliferative properties, mainly attributed to the presence of ellagic acid and p-coumaric acid. It showed strong antiproliferative effects on human cancer cell lines, especially on HL-60 cells, through rapid apoptosis induction. The extract also exhibited antioxidant activity and modulation of cellular oxidative stress and signaling pathways.

Molecular Docking of Natural Compounds for Potential Inhibition of AhR

Summary: The aryl hydrocarbon receptor (AhR) is an important environmental sensor involved in various cellular processes and plays a key role in conditions such as cancer, inflammation, and aging. This study aims to investigate the inhibitory activity of selected natural compounds on AhR. Through computational modeling and docking simulations, additional binding pockets related to AhR inhibition were discovered. In vitro experiments confirmed the efficacy of beta-carotene and ellagic acid in inhibiting AhR activation, validating the computational approach.

A Comparative Study between Microwave Hydrodiffusion and Gravity (MHG) and Ultrasound-Assisted Extraction (UAE): Chemical and Biological Characterization of Polyphenol-Enriched Extracts from Aglianico Grape Pomace

Summary: The wine industry generates a significant amount of grape pomace, which is a waste product that needs proper disposal. Two different technologies, microwave hydrodiffusion and gravity (MHG) and ultrasound-assisted extraction (UAE), were used to extract valuable bioactive compounds from grape pomace. The extracts were further purified through solid-phase extraction (SPE) to obtain three fractions, F1, F2, and F3. It was found that F2-MHG and F3-MHG had phenol contents 6.5 and 8.5 times higher than the non-fractionated extracts, respectively.
