MMP-2 geno-phenotype is prognostic for colorectal cancer survival, whereas MMP-9 is not

The prognostic significance of single-nucleotide polymorphisms (SNPs) and tumour protein levels of MMP-2 and MMP-9 was evaluated in 215 colorectal cancer patients. Single-nucleotide polymorphism MMP-2(-1306T) and high MMP-2 levels were significantly associated with worse survival. Extreme tumour MMP-9 levels were associated with poor prognosis but SNP MMP-9(-1562C>T) was not. Tumour MMP levels were not determined by their SNP genotypes.

The Tango between Cancer-Associated Fibroblasts (CAFs) and Immune Cells in Affecting Immunotherapy Efficacy in Pancreatic Cancer

Summary: The lack of response to therapy in pancreatic ductal adenocarcinoma (PDAC) patients has led to PDAC having a low survival rate. Immunotherapy has shown promise in other cancer types but is ineffective in PDAC due to its unique tumor microenvironment (TME) with low immune activity. Cancer-associated fibroblasts (CAFs) in the TME may play a role in the low immunotherapy response. Understanding the interactions between CAFs and immune cells in the TME could improve immunotherapy for PDAC and similar cancers.

Urokinase-Type Plasminogen Activator Receptor (uPAR) Cooperates with Mutated KRAS in Regulating Cellular Plasticity and Gemcitabine Response in Pancreatic Adenocarcinomas

Summary: Pancreatic ductal adenocarcinomas (PDACs) with urokinase-type plasminogen activator receptor (uPAR) gene amplification or overexpression have a poor prognosis. uPAR reinforces the MEK/ERK signaling pathway and suppresses FAK and CDC42 signaling in tumor cells with a KRAS mutation, promoting cell migration and proliferation, and sensitizing them towards gemcitabine. However, targeting KRAS or uPAR could induce cellular dormancy and make the tumor more resistant to chemotherapy.
