Elimination of the chemotherapy resistant subpopulation of 4T1 mouse breast cancer by haploidentical NK cells cures the vast majority of mice

Metastatic breast cancer is currently incurable despite initial responsiveness, assumingly due to the presence of chemoresistant subpopulations that can be characterized as label retaining cells (LRC). In the 4T1 mouse breast cancer model, we previously achieved cure after Cyclophosphamide and Total Body Irradiation (CY + TBI) followed by haploidentical bone marrow and spleen transplantation (BMSPLT). CY + TBI without transplantation induced only transient impaired tumor growth indicating a critical role of donor immune cells. As it remained unknown if the 4T1 model resembles human disease with respect to the presence of subpopulations of chemoresistant LRC, we now demonstrate this is indeed the case. Chemoresistance of 4T1 LRC was demonstrated by in vitro co-incubation of fluorescently labeled 4T1 cells in limiting dilution with cyclophosphamide, doxorubicin or cisplatinum, after which only LRC containing colonies remained. LRC also remain in vivo after treatment with CY + TBI. Succeeding experiments set up to identify the haploidentical effector cell responsible for cure and, therefore, for the elimination of chemoresistant LRC designate donor NK cells crucial for the anti-tumor effect. NK cell depletion of the haploidentical graft fully abrogated the anti-tumor effect. Increased disease-free survival retained after transplantation of haploidentical bone marrow and NK cell-enriched spleen cell grafts, even in the absence of donor T-cells or of donor bone marrow. Tumor growth analysis indicates the anti-tumor effect being immediate (days). Based on these data, it is worthwhile to explore alloreactive adoptive NK cell therapy as consolidation for patients with metastasized breast cancer.

Summary: IVIG treatment may improve live birth rate in women with recurrent pregnancy loss and underlying immune conditions, especially in subgroups with elevated NK cell percentage and when intervention starts prior to or during conception cycle. However, caution is needed in interpreting these results due to limitations in study design and small sample sizes. Future randomized controlled trials are necessary before widespread clinical use of IVIG.

The Immunomodulatory Role of Hypoxic Tumor-Derived Extracellular Vesicles

Summary: Hypoxia, a characteristic of many cancer types, can suppress the antitumor functions of the adaptive and innate immune system. Tumor-cell-derived extracellular vesicles (EVs), which participate in the communication between tumor cells and immune cells, are also affected by hypoxia and may contribute to immunosuppression. This review summarizes the current understanding of hypoxic cancer-cell-derived EVs in immunosuppression and provides an overview of the factors (miRNA and proteins) enriched in hypoxic tumor-derived EVs and their role in immunomodulation.

Immune Responses 6 Months After mRNA-1273 COVID-19 Vaccination and the Effect of a Third Vaccination in Patients with Inborn Errors of Immunity

Summary: This study investigated the immune responses of 473 patients with inborn errors of immunity (IEI) 6 months after receiving mRNA-1273 COVID-19 vaccines. The results showed that antibody titers and T cell responses in IEI patients also declined after 6 months of vaccination, and were more likely to drop below normal levels compared to healthy individuals. Additionally, the effectiveness of a third mRNA COVID-19 vaccine was limited in patients with common variable immunodeficiency (CVID) who did not seroconvert after previous vaccinations.

Analysis of FCGR3A-p.176Val variants in women with recurrent pregnancy loss and the association with CD16a expression and anti-HLA antibody status

Summary: This study found that the p.Val176Phe polymorphism in the FCGR3A gene is associated with recurrent pregnancy loss. Patients with at least one p.176Val variant showed increased expression of CD16a receptor and anti-HLA antibodies. However, there was no association between FCGR3A-p.176 polymorphism and the presence of class I and class II anti-HLA antibodies.

Summary: This study investigated the levels of serum and intracellular vitamin C in hospitalized COVID-19 patients and found lower serum vitamin C levels in severe and critical cases. The study suggests that intracellular vitamin C levels decrease during COVID-19 infection, while serum levels remain relatively stable. These findings highlight the importance of further research on the role and mechanisms of intracellular vitamin C in COVID-19 infection.
