Nicotine-induced human breast cancer cell proliferation attenuated by garcinol through down-regulation of the nicotinic receptor and cyclin D3 proteins

Previous studies have demonstrated that the persistent exposure of human bronchial epithelial cells to nicotine (Nic) through nicotinic acetylcholine receptors increases cyclin D1 promoter activity and protein expression. The main purpose of this study is to elucidate the carcinogenic role of cyclin D3, which is involved in breast tumorigenesis when induced by Nic. Real-time PCR analysis revealed that cyclin D3 is highly expressed at the mRNA level in surgically dissected breast tumor tissue, compared to the surrounding normal tissue (tumor/normal fold ratio = 17.93, n = 74). To test whether Nic/nicotinic acetylcholine receptor (nAChR) binding could affect cyclin D3 expression in human breast cancer cells, the transformed cell line MCF-10A-Nic (DOX) was generated from normal breast epithelial cells (MCF-10A) with inducible alpha 9-nAChR gene expression, using the adenovirus tetracycline-regulated Tet-off system. Tet-regulated overexpression of alpha 9-nAChR in MCF-10A-Nic (DOX) xenografted BALB/c-nu/nu mice resulted in a significant induction of cyclin D3. In contrast, cyclin D3 expression was down-regulated in alpha 9-nAChR knock-down (siRNA) MDA-MB-231-xenografted tumors in NOD.CB17-PRKDC(SCID)/J(NOD-SCID) mice. Furthermore, we found that Nic-induced human breast cancer (MDA-MB-231) cell proliferation was inhibited by 1 mu M of garcinol (Gar), isolated from the edible fruit Garcinia indica, through down-regulation of alpha 9-nAChR and cyclin D3 expression. These results suggest that alpha 9-nAChR-mediated cyclin D3 overexpression is important for nicotine-induced transformation of normal human breast epithelial cells. The homeostatic regulation of cyclin D3 has the potential to be a molecular target for antitumor chemotherapeutic or chemopreventive purposes in clinical breast cancer patients.

Calebin-A prevents HFD-induced obesity in mice by promoting thermogenesis and modulating gut microbiota

Molecular weight, chain length distribution and long-term retrogradation of cassava starch modified by amylomaltase

Summary: Cassava starch was modified with amylomaltase (AMase) to investigate its structural changes and retrogradation properties. The AMase treatment resulted in an increase in molecular weight and a reorganization of chain distributions in amylose and amylopectin. The extent of retrogradation was influenced by the enzyme/substrate ratio and reaction time. A new insight into the structural characteristics of the modified starch and its long-term retrogradation was gained through the analysis of the long-to-short chain ratio, the enzyme/substrate ratio, and reaction time.

Piceatannol and 3′-Hydroxypterostilbene Alleviate Inflammatory Bowel Disease by Maintaining Intestinal Epithelial Integrity and Regulating Gut Microbiota in Mice

Summary: This study evaluated the effects of Piceatannol (PIC) and 3'-hydroxypterostilbene (HPSB) on DSS-induced colitis in mice. The findings showed that both PIC and HPSB attenuated inflammation by inhibiting specific pathways and reducing inflammasome activation. PIC was also effective in modulating tight junctions. Additionally, both PIC and HPSB altered the composition of gut microbiota. These results suggest that PIC may be more effective in maintaining gut barrier integrity than HPSB and could be a promising ingredient for colitis prevention.

Modulatory Effect of Fermented Black Soybean and Adlay on Gut Microbiota Contributes to Healthy Aging

Summary: This study investigates the anti-aging potential of fermented black soybean and adlay (FBA) supplements. The results show that FBA supplementation prevents muscle loss, reduces aging biomarkers, improves gut microbial dysbiosis, and reverses the reduced ability of microbiota in aged mice. These findings suggest that FBA has significant anti-aging effects and can be developed as a functional food for healthy aging.

Cross-Sectional Study: New Approach for Diagnostic Identification of Non-Robust Older Adult

Summary: This study aims to explore the relationship between serum metabolites and gut microbiota in non-robust older adults as a potential diagnostic biomarker. The results show significant differences in gut microbial composition between robust and non-robust subjects, particularly with the abundance of Escherichia/Shigella. Moreover, the abundance of Escherichia/Shigella is positively correlated with the levels of discriminant metabolites. These findings indicate the clear interrelation between gut microbiota and serum metabolites in non-robust older adults, suggesting Escherichia/Shigella as a potential biomarker candidate for robustness sub-phenotypic identification.

Prevention of Glutamate-Induced Neurodegeneration by Piceatannol via Mitochondrial Rescue In Vitro and In Vivo

Summary: This study aims to evaluate the preventive effect of piceatannol on glutamate-induced neurodegeneration via mitochondrial rescue. The results show that piceatannol can prevent glutamate-induced apoptosis, reduce mitochondrial reactive oxygen species accumulation, promote mitochondrial biogenesis, and preserve mitochondrial functionality. Piceatannol effectively protects dopaminergic neurons from degradation and preserves their controlled responses.

Bioactives from Crude Rice Bran Oils Extracted Using Green Technology

Summary: Crude rice bran oils from different rice cultivars and extraction methods have varying levels of nutraceuticals. The health benefits of rice bran oil in lowering cholesterol are due in part to non-nutrient nutraceuticals such as ?-oryzanol, phytosterols, and policosanols. In this study, the nutraceutical composition and antioxidant properties of crude rice bran oils extracted using different technologies were compared. The results showed that subcritical liquefied dimethyl ether extraction produced oils with higher yield and higher levels of phenolic compounds, ?-oryzanol, phytosterols, and policosanols compared to conventional hexane extraction and supercritical CO2 extraction. Overall, the choice of extraction method significantly affected the composition and properties of the crude rice bran oil.

The Role of Thrombomodulin in Estrogen-Receptor-Positive Breast Cancer Progression, Metastasis, and Curcumin Sensitivity

Summary: This study aims to evaluate the role of thrombomodulin (TM) in ER-positive (ER+) breast cancer. The results showed that lower TM expression is associated with poor overall survival and relapse-free survival rates in ER+ breast cancer patients. Silencing TM in MCF7 cells increased cell proliferation, migration, and invasion ability, while overexpression of TM in T47D cells led to decreased cell proliferation, migration, and invasion ability. Furthermore, TM regulates curcumin sensitivity by interfering with the expression of drug-resistant genes.

CWH43 Is a Novel Tumor Suppressor Gene with Negative Regulation of TTK in Colorectal Cancer

Inhibition of CWH43 promotes cancer cell proliferation, migration, and invasiveness, while its overexpression has inhibitory effects. CWH43 appears to regulate the TTK-mediated cell cycle, impacting the development of colorectal cancer.

The therapeutic potential of curcumin and its related substances in turmeric: From raw material selection to application strategies

Summary: Turmeric is a medicinal plant commonly used in Chinese and Indian traditional medicine. It has various health benefits due to the active components called curcuminoids. This article reviews the composition of turmeric and the properties of curcumin, highlighting its antioxidant, anti-inflammatory, anti-diabetic, anti-colorectal cancer, and other physiological activities. It also discusses the challenges of using curcumin and provides three novel application strategies to overcome these limitations.
