Summary: In this study, the effects of a new Ag(I)curcuminoid-Gboxin analog complex were investigated using the MCF-7 breast carcinoma cell line. The results showed that Ag(I)FLLL49-GbA could up-regulate the expression of caspase-3, P53, and Bax in MCF-7 cells after 48 hours of treatment. However, it was puzzling because MCF-7 cells do not naturally express caspase-3. Appropriate modifications are suggested to strengthen the findings of this study.

Summary: This study examined the effects of TaPcZn-PDT on cytotoxicity, cell viability, apoptosis, pyroptosis, cellular reactive oxygen species (ROS), mitochondrial membrane potential (??m), caspase-1, caspase-3, and nuclear transcription factor-?B (NF?B) in MCF-7 cells. The results showed that TaPcZn-PDT increased cytotoxicity and the percentage of apoptotic and pyroptotic cells, decreased cell viability and ??m, induced ROS production, and activated caspase-1, caspase-3, and NF?B in MCF-7 cells. siRNA targeting caspase-1 attenuated the effects of TaPcZn-PDT on apoptosis, pyroptosis, and the activation of caspase-1, caspase-3, and NF?B in MCF-7 cells. Therefore, caspase-1 plays a regulatory role in the apoptosis and pyroptosis induced by TaPcZn-PDT in MCF-7 cells.

Summary: Breast cancer MCF-7 cell-line-derived mammospheres exhibit enriched CD44+/CD24- cells resembling breast cancer stem cells, expressing high levels of S1P3. Growth-promoting agents induce SphK1 and S1P3 translocation to the nucleus, while TNF alpha inhibits their nuclear translocation and promotes apoptosis in mammospheres. The study suggests inhibiting SphK1 and S1P3 nuclear translocation as a novel approach to prevent BCSC proliferation.

Summary: In this study, an efficient chemosensor (PCF) based on the pyridine-carbazole moiety was developed for the differential detection of aspartic acid in biological systems. PCF showed strong binding affinity towards aspartic acid, with a nanomolar detection limit. The binding stoichiometry of aspartic acid and PCF was determined to be 1:1 from Jobs plot analysis. Furthermore, the efficacy of PCF was successfully demonstrated in in vitro experiments in MCF-7 breast cancer cells.

Summary: Carbazole compounds PK9320 and PK9323, as second-generation analogues of PK083, have been shown to restore p53 signaling in cancer cells by binding to mutation-induced surface crevices. The study demonstrates that small modifications in carbazole substitution patterns can significantly impact their genotoxic and epigenetic properties, with PK9320 and PK9323 being potential candidates for anticancer and epi-compounds.

Summary: This study evaluated the cytotoxicity and toxicological effect of methanol fruit extracts of Artoarpus altilis against MCF-7 cells. The methanol extract of the pulp part showed the strongest inhibition concentration on MCF-7 cells. Furthermore, the methanol extracts induced apoptosis and cell cycle arrest in a time-dependent manner by regulating the expression of apoptosis-related genes, suggesting their potential anticancer activity.

Summary: This study aimed to demonstrate that IGFBP-3 induces cellular senescence and inhibits cell proliferation in MCF-7 breast cancer cells by suppressing telomerase activity. The results showed that IGFBP-3 induction led to loss of cell viability, increased percentage of non-cycling cells, morphological alterations associated with senescence, and decreased telomerase activity. These findings suggest that IGFBP-3 is a negative regulator of MCF-7 breast cancer cell growth through senescence induction and telomerase suppression.

Summary: A new Sn(IV) tetraarylchlorin with thien-2-yl substituents (SnC) has been synthesized and characterized, showing high Phi(Delta) value and promising photodynamic activity against MCF-7 breast cancer cells. These results suggest that Sn(IV) tetraarylchlorins like SnC are suitable candidates for further photodynamic therapy studies.

Summary: Radiation therapy is a crucial part of cancer treatment, but it has adverse effects. Melatonin, as an adjuvant molecule, can limit damage to normal tissues and enhance the efficacy of radiation therapy in breast cancer. This study revealed that melatonin can potentiate the anti-proliferative effect of radiation, modulate gene and microRNA expression, and enhance the inhibition of cancer growth. Melatonin shows promise as both a radiosensitizer and radioprotector in breast cancer treatment.

Summary: This study investigates the effects of severe hypoxia, site-specific inhibition of respiratory chain components, and uncouplers on 2D-cultured HepG2 and MCF-7 tumor cells. The results show that there is no correlation between oxidative phosphorylation activity and viability in these cell lines, indicating that the connection between oxidative phosphorylation and cancer cell survival is dependent on cell type and conditions.
