Summary: This study evaluated two commonly used automated computed tomography perfusion software applications and found that by adjusting the relative cerebral blood flow thresholds, comparable results with reliable information on the follow-up infarct volume after complete recanalization can be obtained. Keeping the software specific default settings means being more inclusive in patient selection but sacrificing the highest possible accuracy in the estimation of the infarct volume.

Summary: Stroke is caused by ruptured or blocked cerebral blood vessels and subsequent disruption of brain blood circulation, resulting in rapid neurological deficits. The majority of stroke cases are ischemic strokes. Current treatments for ischemic stroke, such as t-PA thrombolytic therapy and surgical thrombectomy, can paradoxically lead to ischemia-reperfusion injury, exacerbating brain damage. Minocycline, a semi-synthetic tetracycline antibiotic, has demonstrated a wide range of neuroprotective effects independent of its antibacterial activity. In this article, we summarize the mechanisms underlying the protective effects of minocycline against cerebral ischemia-reperfusion injury, including modulation of oxidative stress, inflammatory response, excitotoxicity, programmed cell death, and blood-brain barrier injury. We also discuss the role of minocycline in alleviating stroke-related complications, providing a theoretical basis for its clinical application in cerebral ischemia-reperfusion injury.

Summary: This study reveals that baicalin, derived from Scutellaria baicalensis, can protect lung alveolar epithelial cells from H1N1-induced pyroptosis, an inflammatory form of programmed cell death, by inhibiting the caspase-3/GSDME pathway. This is the first demonstration of the important role of baicalin in antiviral activity.

Summary: Despite significant progress in stroke therapy, ischemic stroke still imposes a high socioeconomic burden on industrialized countries. Current treatment paradigms focus on thrombolysis or thrombectomy, which are only suitable for a fraction of patients. Therefore, there is an urgent need for adjuvant therapeutic approaches like neuroprotection.

Summary: This study showed that baicalin protects neurons from oxidative stress, inflammation, and apoptosis by promoting astrocytes to release more BDNF. The neuroprotective effect is mainly mediated through the BDNF-TrkB pathway and downstream signaling regulators PI3K/Akt and MAPK/ERK1/2.

Summary: Cerebral ischemia leads to excitotoxicity and neural apoptosis. In this study, the effects of chemogenetic inhibition or activation of glutamatergic neurons in the vCA1 region on sequelae induced by cerebral ischemia were investigated. The results showed that inhibition of vCA1 pyramidal neurons alleviated neurological deficits, pain perception, anxiety, and depression, while activation had limited effects. Additionally, overexpression of CREB protein in vCA1 pyramidal neurons improved the dysfunction induced by ischemic stroke by upregulating BDNF.

Summary: Combination therapy with increased efficacy and reduced toxicity is crucial for treating complex diseases. This study presents a strategy called Driver-induced Modular Screening (DiMS) to elucidate the additive mechanisms of Baicalin (BA) and Jasminoidin (JA) on cerebral ischemia (CI) therapy. The study identifies driver genes and modules and explores their neuroprotective roles. The findings provide new insights into the synergistic mechanisms of combination therapy and screening multi-targeted drugs for complex diseases.

Summary: This study optimized a nanocarrier formulation for nose-to-brain delivery of neuroprotective agents. The results showed that the nanocarriers could effectively deliver the agents to the brain, reducing delivery to the peripheral circulation. By modifying the surface of the nanoparticles, the bioavailability of the agents was improved, and neuroprotective effects against cerebral ischemia were observed.
