Summary: Mesenchymal stem cells (MSCs) have been used in clinical studies due to their versatility and low immunogenicity. However, safety issues remain a concern. Recently, extracellular vesicles (EVs) derived from MSCs have been explored as an alternative. EVs are nano-sized particles released by cells and have similar biological functions as their origin cells. They carry bioactive molecules and facilitate tissue regeneration. EVs derived from adipose-derived MSCs (ADMSC) also have neuroprotective and neurogenesis effects. This study aimed to enhance the neural differentiation ability of ADMSC-derived EVs by investigating the neurogenic-differentiation process. The results showed that differentiated EVs (dEVs) obtained from neurogenesis conditioned media increased neurogenic ability by altering microRNA expression and cytokine composition. dEVs promoted neuronal differentiation of neural progenitor cells in vitro, indicating their potential in EV-based therapy for neurological disorders.

Summary: Autism spectrum disorder (ASD) involves enhanced processing of amyloid-beta precursor protein (APP), leading to increased levels of sAPP alpha in the blood and accumulation of N-terminally truncated A beta peptides in the brain cortex and subcortical structures, particularly in GABAergic neurons expressing parvalbumin. A beta accumulation is also observed in epilepsy, a common co-morbidity of ASD. A beta peptides can induce electroconvulsive episodes. Traumatic brain injuries resulting from self-injurious behaviors, another co-morbidity of ASD, frequently result in increased production and altered processing of APP as well as accumulation of A beta in the brain. The consequences of A beta accumulation in neurons and synapses vary depending on the A beta species, modifications, concentration, aggregation, and location. These effects include modulation of transcription, induction of oxidative stress, alteration of membrane receptors' signaling, calcium channel formation causing hyper-activation of neurons, and reduction of GABAergic signaling, all of which disrupt synaptic and neuronal network functions. In conclusion, ASD, epilepsy, and self-injurious behaviors contribute to the increased production and accumulation of A beta peptides, which in turn contribute to the dysfunctions of neuronal networks manifested as autism symptoms, epilepsy, and self-injurious behaviors.
