Summary: The treatment of chronic myeloid leukemia has improved with the use of tyrosine kinase inhibitors; however, the prognosis for CML in the blast phase remains poor and treatment options are limited. In order to provide consistent therapy for children and adolescents with this high-risk disease, an international panel of pediatric CML experts developed recommendations based on current literature and national standards.

Differential expression of miRNAs from extracellular vesicles in chronic graft-versus-host disease: A preliminary study

Summary: This study aimed to identify biomarkers associated with the occurrence of cGvHD by analyzing miRNAs in plasma EVs. The results revealed that hsa-miR-630 and hsa-miR-374b-5p were downregulated in cGvHD patients, while hsa-miR-29c-3p was upregulated.

Summary: This study retrospectively evaluated the clinical course and test results of 7 patients diagnosed with ADA deficiency in Poland from 2010 to 2022. The patients demonstrated immune and non-immune symptoms, and 5 of them underwent hematopoietic stem cell transplantation. Early diagnosis is crucial to prevent irreversible multi-organ failure.

The impact of NKG2A and NKG2D receptors and HLA-E and MICA ligands polymorphisms on post-transplant complications after paediatric allogeneic HSCT: a single-centre experience

Summary: This study investigated the impact of NKG2 receptors and their ligands polymorphisms on post-transplant complications after haematopoietic stem cell transplantation (HSCT) in children. The HLA-E rs1264457 polymorphism was significantly associated with cytomegalovirus (CMV) infection. NKG2A or MICA polymorphisms showed a strong trend towards the risk of acute graft-versus-host disease (GvHD). These findings suggest that genotyping results may have prognostic value for the outcome of paediatric HSCT.

Segmental chromosomal aberrations as the poor prognostic factor in children over 18 months with stage 3 neuroblastoma without MYCN amplification

Summary: Patients with stage 3 neuroblastoma without MYCN amplification show heterogeneity in disease presentation and prognosis. Age at diagnosis, INPC diagnostic category, chromosomal aberrations, and biochemical markers are prognostic factors. The risk of treatment failure is higher in patients with segmental chromosomal aberrations, especially in those over 18 months, and may require more intensive treatment.

Quantitative and Qualitative Airborne Mycobiota Surveillance in High-Risk Hospital Environment

Summary: The primary aim of this study was to assess the prevalence of fungi in selected hospital wards and evaluate the susceptibility of Aspergillus fumigatus isolates to triazoles. Air samples were collected and cultured on Sabouraud agar. The study found that protected rooms with sterile air circulation and air disinfection devices had significantly lower amounts of cultured fungi compared to unprotected rooms. The most contaminated areas were corridors and bathrooms, with Cladosporium and Penicillium being the dominant species. A rare outbreak of A. fumigatus spores was noted in a hospital for lung diseases but no triazole-resistant isolates were detected. Regular microbiological testing of the hospital environment can help detect spore outbreaks and enable corrective procedures.

Summary: Venetoclax has shown significant clinical activity in relapsed or refractory leukemias in paediatric patients in Poland. Although not currently incorporated into paediatric treatment schedules, it has been used in patients who have failed conventional therapy. This study aims to gather clinical data and correlates of paediatric patients treated with Venetoclax to guide its clinical use and stimulate further research.

Serum miRNA-based signature indicates radiation exposure and dose in humans: A multicenter diagnostic biomarker study

Summary: The study demonstrates that serum miRNAs can be used as functional biodosimeters to predict the biological impact of radiation doses in humans undergoing total body irradiation. A logistic regression model was built using miR-150-5p, miR-30b-5p, and miR-320c, which showed high accuracy in identifying samples drawn after irradiation.

Summary: A retrospective study conducted in Poland demonstrated the feasibility and benefits of using Dinutuximab beta for maintenance treatment in patients with high-risk neuroblastoma. Priapism at Diagnosis of Pediatric Chronic Myeloid Leukemia: Data Derived from a Large Cohort of Children and Teenagers and a Narrative Review on Priapism Management

Summary: Pediatric chronic myeloid leukemia (CML) is a rare malignancy with priapism observed in approximately 3.2% of boys with pediatric CML. Priapism duration varied from hours to months, with intermittent priapism being the most common presentation. All patients exhibited splenomegaly and massive leukocytosis. Interventions for priapism included intravenous fluids, heparin, penile puncture, sympathomimetics, and surgical shunt operations. Long-term survivors responded well to treatment and maintained full erectile function.

Summary: Mutations in the FLT3 gene are associated with poor outcomes in pediatric AML. A retrospective analysis of a nationwide pediatric AML database showed that FLT3-ITD-positive patients had worse survival compared to FLT3-ITD-negative patients. Treatment protocols and the use of FLT3 inhibitors and stem cell transplantation showed potential for improving outcomes. Additional genetic mutations, such as WT1 and NPM1, significantly influenced prognosis.

Physical Activity Level and Quality of Life of Children Treated for Malignancy, Depending on Their Place of Residence: Poland vs. the Czech Republic: An Observational Study

Summary: This study aimed to assess the level of physical activity (PA) and quality of life of cancer-treated children, depending on their place of residence (Poland vs. the Czech Republic, where incidence and mortality rates of childhood malignancies are similar). The study showed significant differences in PA levels between Polish and Czech children. Physically active children engaging in more frequent PA reported higher physical performance, higher energy levels, and less mood disturbance. Our study emphasized the importance of promoting tailored PA programs for cancer-treated children to improve well-being and quality of life.
