Summary: This study identified the epigenetic mechanisms underlying the reduction of 4.1N/EPB41L1 in NSCLC. The promoter region of the 4.1N/EPB41L1 gene was highly methylated in LUAD and LUSC patients. Higher methylation levels in the 4.1N/EPB41L1 gene promoter were associated with shorter overall survival in LUAD patients and longer overall survival in LUSC patients. The study also found that the expression of miR-454-3p was abnormally high in NSCLC and directly targeted the 4.1N/EPB41L1 mRNA. The findings suggest that promoter hypermethylation and abnormal miR-454-3p expression restrict the expression of 4.1N/EPB41L1 in NSCLC.

Summary: Objective: This study screened core genes in lung adenocarcinoma (LUAD) using bioinformatics, evaluated their prognosis value and role in immune cell infiltration. Methods: Gene chips from GEO database were analyzed to obtain differentially expressed genes, and then further analyzed using DAVID, Cytoscape, and PPI network analysis. Several databases were used for expression verification, survival analysis, and immune infiltration relationship identification. CCNB2 and CDC20 were identified as vital core genes. Conclusion: CCNB2 and CDC20 are essential genes that may serve as prognostic biomarkers in LUAD and play a role in immune infiltration and protein expression processes.

Summary: DriverSubNet is an effective parameter-free algorithm for detecting driver genes in cancers by mining mutation and gene expression information. Compared to classic methods, DriverSubNet can rank driver genes and filter out passenger genes more efficiently, identifying both known and unknown cancer driver genes that could serve as potential therapeutic targets and useful prognostic biomarkers for cancer patients. Therefore, DriverSubNet may serve as a valuable tool for identifying driver genes through subnetwork enrichment analysis.

Summary: This study identified three molecular subtypes of lung adenocarcinoma (LUAD) based on necroptosis-related genes. The C3 subtype showed better prognosis and higher immune cell infiltration. The C1 subtype had poor prognosis and higher clinical grade, with a high mutation frequency in TP53 gene. Gene set enrichment analysis revealed aberrant cell cycle-related pathways in both C1 and C3 subtypes. A risk model based on necroptosis phenotype-related genes was constructed, showing strong ability to predict survival.

Summary: Modulation of brain olfactory (OR) and taste receptor (TASR) expression was found in neurological diseases, such as Alzheimer's disease (AD). In this study, the expression and regulation of OR and TASR were explored in the human orbitofrontal cortex (OFC) of sporadic AD patients. The results showed that the genes were downregulated at early stages of AD, suggesting transcriptional regulation through epigenetic mechanisms. The interaction between H3K9me3 and MeCP2 was also discovered, implicating MeCP2 in the transcriptional regulation of OR and TASR in AD.
