Sensitive Detection of Single-Nucleotide Mutation in the BRAF Mutation Site (V600E) of Human Melanoma Using Phosphate-Pyrene-Labeled DNA Probes

A series of novel nucleotide phosphoramidites were rationally designed and synthesized and were then site-specifically incorporated in DNA oligonucleotide probes with pyrene-modified phosphate. These oligodeoxynucleotide (ODN) probes almost have no inherent fluorescence emission with pyrene modification at 3' phosphate of corresponding nucleotides as a result of the photoinduced electron-transfer quenching effect of nudeobases (thymidine similar to cytidine > guanosine >> adenosine). However, strong fluorescence emission was observed only with the perfectly matched duplex for the probes with pyrene modified at 3' phosphate of thymidine and cytidine. These rationally designed ODN probes successfully worked as turn on fluorescence oligonucleotide sensors for single-nucleotide polymorphism (SNP) and were used for detecting a single BRAF mutation site (V600E) of human melanoma.

Smart Biomaterials for Articular Cartilage Repair and Regeneration

Summary: Articular cartilage defects cause disability and global socioeconomic loss. However, the limited regeneration ability of cartilage tissue due to its avascular and nearly acellular nature poses challenges for repair and regeneration. Smart biomaterials can regulate cell behavior, provide a favorable microenvironment and induce articular cartilage repair and regeneration. The use of smart drug release systems and smart scaffolds in these processes is discussed, as well as the importance of advanced manufacturing techniques and clinical translation of smart biomaterials.

Ionic liquid-based aggregation towards the mesoscale mechanism and their applications

Summary: This Perspective discusses the structures of ionic liquids (ILs) and the aggregations that can significantly alter their micro-environment and impact their properties and functions. The mesoscale structures of IL aggregation in pure ILs and ILs-solvent mixtures are analyzed based on interacting forces and thermodynamics. The potential applications of IL aggregation in various scientific and engineering fields are explored, such as biomass conversion, smart materials, and life medicine. The challenges and opportunities in the field of IL aggregation, including direct observation techniques, large-scale simulations, theoretical forming mechanisms, and coupling mechanisms with solid interfaces, are also discussed. These new perspectives shed light on the rational design of ILs and generate research interest in ILs as a green medium in chemical engineering.

Purification identification and function analysis of ACE inhibitory peptide from Ulva prolifera protein

Summary: In this study, Ulva prolifera, an edible alga, was used to prepare angiotensin-I converting enzyme (ACE) inhibitory peptide. The highest ACE inhibitory activity was obtained from the hydrolysate of algae protein, which was processed using various methods and identified as the peptide DIGGL. DIGGL exhibited non-competitive inhibition against ACE and mainly achieved this effect through three conventional hydrogen bonds. It also showed the ability to activate endothelial nitric oxide synthase and decrease endothelin-1 secretion in human umbilical vein endothelial cells. Moreover, DIGGL promoted splenocyte proliferation and remained active during gastrointestinal protease digestion.

Physicochemical, structure properties and in vitro hypoglycemic activity of soluble dietary fiber from adlay (Coix lachryma-jobi L. var. ma-yuen Stapf) bran treated by steam explosion

Summary: This study investigates the effects of steam explosion (SE) treatment on the physicochemical, structural properties, and in vitro hypoglycemic activities of adlay bran soluble dietary fiber (SDF). The results show that SE treatment significantly increases the SDF content and improves its functional properties. The SE-treated SDF exhibits higher oil-holding capacity, cholesterol adsorption capacity, glucose adsorption capacity, glucose dialysis retardation index, and alpha-amylase activity inhibition ratio compared to the SDF from untreated adlay bran.

Simultaneous analysis of the oxidation of solvent-extracted and cold-pressed green coffee oil during accelerated storage using 1H NMR and 13C NMR spectroscopy

Summary: This study investigated the oxidation status of solvent-extracted and cold-pressed green coffee oil (GCO) under accelerated storage conditions using proton nuclear magnetic resonance (1H and 13C NMR) spectroscopy. Results showed that the signal intensity of oxidation products gradually increased with oxidation time, while unsaturated fatty acid signals weakened. The NMR system was found to be a fast and convenient tool for monitoring the oxidation process and quality control of GCO.

Novel class of crown ether functionalized ionic liquids with multiple binding sites for efficient separation of lithium isotopes

Summary: In this study, a novel class of functionalized ionic liquids with grafted crown ether-moieties (CE-FILs) were reported for the efficient separation of Li-6 and Li-7. The enhanced distribution of Li+ was achieved by the synergistic effect of ion-dipole interaction, hydrogen bonding, and coordination interaction. The CE-FILs pave a new way for Li isotopes separation.

Quantitative Relation between Ionic Diffusivity and Ionic Association in Ionic Liquid-Water Mixtures

Summary: Molecular dynamic simulations were conducted to investigate the relationship between the microscopic structures changed by water and the ionic diffusivity in aqueous mixtures of imidazolium ionic liquids (ILs). Two distinct regimes of ionic diffusivity were identified with increasing water concentrations, which are directly correlated to the ionic association. The study also revealed two general relationships between ionic diffusivity and ion-pair lifetimes, as well as short-ranged interactions between cations and anions.

Summary: The control of DNA assembly systems on cells is crucial for precise targeted therapies. In this study, a controllable DNA self-assembly system based on the regulation of G-quadruplex DNA topology by a reduction-sensitive azobenzene ligand is reported. The ligand can induce a complete change of G4 topology in a tissue-specific manner, allowing control of the self-assembly of G4-functionalized DNAs on cells.

One-step synthesis of novel carbon dots through cross amide formation of benzene-1,3,5-triamine and trimethyl 1,3,5-benzenetricarboxylate for fluorescence Hg2+ detection

Summary: Novel green-emitting fluorescent carbon dots (CDs) were successfully designed and synthesized through cross amide formation of two precursors under hydrothermal treatment. The obtained CDs exhibited good photoluminescent properties with a high quantum yield. They also demonstrated good fluorescence sensitivity and selectivity for Hg2+, making them applicable for Hg2+ detection in real water samples and intracellular Hg2+ fluorescence detection.

Electron Density Learning of Z-Bonds in Ionic Liquids and Its Application

Summary: In this study, a machine learning model targeting the electronic density of Z-bonds in ionic liquids (ILs) was successfully trained. The correlations between the Z-bond energy and the electrochemical properties of IL-based systems were investigated separately. This model provides an efficient way to predict the electronic density of Z-bonds in nanoscale systems and offers insights into the electrochemical properties of ILs.

Cathepsin B-activatable cyclic antisense oligonucleotides for cell-specific target gene knockdown in vitro and in vivo

Light-triggered site-directed RNA editing by endogenous ADAR1 with photolabile guide RNA

Summary: RNA A-to-I editing is a common post-transcriptional modification in cells. By using guide RNA and exogenous ADAR enzymes, artificial intervention of A-to-I editing can be achieved at specific RNA sites. In this study, we developed photo-caged antisense guide RNA oligonucleotides with a simple cholesterol modification and successfully achieved light-triggered site-specific RNA A-to-I editing for the first time using endogenous ADAR enzymes. Our caged A-to-I editing system effectively manipulated RNA editing in living cells and 3D tumorspheres, providing a new approach for precise manipulation of gene expression and spatial regulation.
