Use of in vivo-induced antigen technology (IVIAT) for the identification of Streptococcus suis serotype 2 in vivo-induced bacterial protein antigens

Background: Streptococcus suis serotype 2 (SS2) is a zoonotic agent that causes death and disease in both humans and swine. A better understanding of SS2-host molecular interactions is crucial for understanding SS2 pathogenesis and immunology. Conventional genetic and biochemical approaches used to study SS2 virulence factors are unable to take into account the complex and dynamic environmental stimuli associated with the infection process. In this study, in vivo-induced antigen technology (IVIAT), an immunoscreening technique, was used to identify the immunogenic bacterial proteins that are induced or upregulated in vivo during SS2 infection. Results: Convalescent-phase sera from pigs infected with SS2 were pooled, adsorbed against in vitro antigens, and used to screen SS2 genomic expression libraries. Upon analysis of the identified proteins, we were able to assign a putative function to 40 of the 48 proteins. These included proteins implicated in cell envelope structure, regulation, molecule synthesis, substance and energy metabolism, transport, translation, and those with unknown functions. The in vivo-induced changes in the expression of 10 of these 40 genes were measured using real-time reverse transcription (RT)-PCR, revealing that the expression of 6 of the 10 genes was upregulated in the in vivo condition. The strain distribution of these 10 genes was analyzed by PCR, and they were found in the most virulent SS2 strains. In addition, protein sequence alignments of the newly identified proteins demonstrate that three are putative virulence-associated proteins. Conclusion: Collectively, our results suggest that these in vivo-induced or upregulated genes may contribute to SS2 disease development. We hypothesize that the identification of factors specifically induced or upregulated during SS2 infection will aid in our understanding of SS2 pathogenesis and may contribute to the control SS2 outbreaks. In addition, the proteins identified using IVIAT may be useful potential vaccine candidates or virulence markers.

Chemosensor with Ultra-High Fluorescence Enhancement for Assisting in Diagnosis and Resection of Ovarian Cancer

Summary: Fluorescence imaging-guided diagnostics is a promising method for detecting tumors in situ due to its simplicity and non-invasiveness. The chemosensor LAN-beta gal, with a specific recognition unit of d-galactose residue and a fluorophore of LAN-OH, showed remarkable fluorescence enhancement after beta-gal was digested. It successfully visualized endogenous beta-gal, distinguished ovarian cancer cells from normal cells, and was applied in vivo for tumor detection and surgical resection of ovarian cancer. The LAN-beta gal chemosensor shows potential as a new tool for the clinical diagnosis and treatment of ovarian cancer.

Summary: In this study, optimal ultrasound pretreatment was used to extract lignin and hemicellulose from recalcitrance-reduced rice mutant straw to improve cellulose accessibility. Intermittent ultrasound-assistant enzymatic hydrolyses were then conducted to increase hexose yield and bioethanol concentration. This study also generated biochar with high porosity and strong adsorption capacity for dyes. Therefore, this research demonstrates a green process technology for high-yield bioethanol and high-porosity biochar from lignocellulose.

Effect of melatonin on ATG2B-mediated autophagy regulation in sheep granulosa cells with different FecB genotypes

Summary: The aim of this study was to investigate the molecular mechanisms underlying MLT regulation of autophagy in GCs with different Fec(B) genotypes. The researchers found that autophagy levels were significantly higher in GCs with Fec(B) BB genotype than in those with Fec(B)++ genotype. ATG2B was associated with cell autophagy and highly expressed in GCs with Fec(B) BB genotype. MLT could protect GCs by decreasing ROS levels, especially in GCs with Fec(B) ++ genotype.

Effects of Enterobacter sp. Zm-123 and spent mushroom substrate on rape growth promotion and cadmium toxicity reduction in cadmium-contaminated soil

Summary: The study investigates the effects of plant growth-promoting rhizobacteria (PGPR) Enterobacter sp. Zm-123 and a heavy metal adsorption biomass spent mushroom substrate (SMS) on the growth promotion of rape and reduction of cadmium (Cd) toxicity in contaminated soil. Results show that all treatment groups significantly improve soil properties and decrease exchangeable Cd (SE-Cd) in the soil. Additionally, the treatments increase the biomass and chlorophyll content of rape, enhance antioxidant enzyme activities, reduce proline and malondialdehyde (MDA) accumulation, and decrease the Cd content in the edible parts of rape. Among the treatments, the semi-solid fermentation (SSF) group demonstrates the strongest effect.

Comparing carbon nanoparticles and indocyanine green for sentinel lymph node mapping in endometrial cancer: A randomized-controlled single-center trial

Summary: This study compared the feasibility and diagnostic value of sentinel lymph node (SLN) mapping using carbon nanoparticles (CNPs) and indocyanine green (ICG) in endometrial cancer (EC). The results showed that the detection rate, sensitivity, and negative predictive value were similar between the two methods. The study suggested that CNPs could be an alternative to ICG for SLN mapping when near-infrared imaging equipment is lacking.

Tunable Hierarchically Structured Meso-Macroporous Carbon Spheres from a Solvent-Mediated Polymerization-Induced Self-Assembly

Summary: In this study, a versatile solvent-mediated polymerization-induced self-assembly (PISA) strategy was developed for the direct synthesis of highly N-doped hierarchically porous carbon spheres with tunable meso-macroporous configuration. The introduction of intermolecular hydrogen bonds enhanced the interfacial interactions between block copolymers, oil droplets, and polyphenols. Different cosolvent systems were used to systematically manipulate the dominant hydrogen-bond-driven interactions, resulting in diverse porous structures. The hierarchically structured meso-macroporous N-doped carbon spheres exhibited tunable sphere sizes and mesopores and macropores, providing a new platform for the synthesis of well-defined porous materials.

Irreversible biosynthesis of D-allulose from D-glucose in Escherichia coli through fine-tuning of carbon flux and cofactor regeneration engineering

Summary: By modifying the glycolytic pathway of Escherichia coli and optimizing the whole-cell reaction condition, the engineered strain achieved high-yield production of d-allulose from low-cost substrate. The cell growth inhibition was alleviated and d-allulose production increased by developing an ATP regeneration system for enhanced substrate phosphorylation. Fine-tuning of carbon flux and implementation of fed-batch co-substrate strategy further improved the d-allulose titer and conversion rate.

Utilizing the influence of multiple potential factors for social recommendation

Summary: The recommendation system helps users make rational decisions by assisting them in selecting satisfactory products and services. To enhance recommendation accuracy, social information has been incorporated into most methods in recent years. However, current social recommendations only consider the impact of historical items and social information on users. This study proposes a model called Utilizes the influence of multiple potential factors for Social Recommendation (UimSRec) to capture the influence of various potential factors on users, including consumption experience, life cycle, social consumption culture, and social roles. By simulating these influences in the form of latent semantics and associating them with user representation, UimSRec improves recommendation performance significantly through modeling diverse potential impact information and their relationships.

Fast Predicting the Complex Nonlinear Dynamics of Mode-Locked Fiber Laser by a Recurrent Neural Network with Prior Information Feeding

Summary: A recurrent neural network-based Fourier decomposition method is proposed for fast and accurate modeling of femtosecond mode-locked fiber lasers. Generalization over different cavity settings is achieved through a proposed prior information feeding method. With GPU acceleration, the mean time for the AI model to infer 500 roundtrips is less than 0.1 s, which is approximately 146 times faster than the traditional method. This AI-enabled approach is promising to become a standard method for femtosecond laser modeling.

Functionally integrated self-healable and redox bromide-ion additive hydrogel electrolyte for Zn-ion hybrid supercapacitors

Summary: Functional hydrogel electrolytes are essential for functional energy storage devices, and their performance relies on the functional groups and interactions within the polymer chain. This study successfully synthesized a self-healable and redox bromide-ion additive hydrogel electrolyte (SAM-Zn) using a natural polymer backbone. The SAM-Zn showed remarkable self-healing ability, excellent mechanical properties, and high energy density in Zn-ion hybrid supercapacitors. This work provides a simple strategy for combining self-healing properties and excellent electrochemical performance in hydrogel electrolytes.

Cochlear implantation in a patient with congenital microtia, cochlear hypoplasia, venous anomalies of the temporal bone and laryngomalacia: Challenges and surgical considerations

Summary: This study reports the case of a 28-month-old girl with congenital profound hearing loss, laryngomalacia, and malformed inner ear, who received cochlear implantation surgery. The genetic basis was determined using trio-based whole exome sequencing, and perioperative evaluation and management of the airway was performed by an experienced anesthesiologist. Cochlear implantation was eventually performed successfully under uneventful general anesthesia.

3D Macrocyclic Structure Boosted Gene Delivery: Multi-Cyclic Poly(β-Amino Ester)s from Step Growth Polymerization

Summary: The topological structures of polymers are crucial for determining their efficiency in gene delivery. This study synthesized a new generation of multi-cyclic poly(& beta;-amino ester)s (CPAEs) with unique topology structure for the first time via step growth polymerization. Through controlling the occurrence stage of cyclization, three types of CPAEs with rings of different sizes and topologies were obtained. In vitro experiments showed that the CPAEs with macro rings (MCPAEs) significantly enhanced transgene expression compared to their branched counterparts. Additionally, the MCPAE vector with optimized terminal group efficiently delivered the CRISPR plasmid encoding Staphylococcus aureus Cas9 nuclease and dual guide sgRNAs for gene editing therapy.
