Association of HPA axis-related genetic variation with stress reactivity and aggressive behaviour in pigs

Background: Stress, elicited for example by aggressive interactions, has negative effects on various biological functions including immune defence, reproduction, growth, and, in livestock, on product quality. Stress response and aggressiveness are mutually interrelated and show large interindividual variation, partly attributable to genetic factors. In the pig little is known about the molecular-genetic background of the variation in stress responsiveness and aggressiveness. To identify candidate genes we analyzed association of DNA markers in each of ten genes (CRH g. 233C>T, CRHR1 c.*866_867insA, CRHBP c.51G>A, POMC c.293_298del, MC2R c.306T>G, NR3C1 c.*2122A>G, AVP c.207A>G, AVPR1B c.1084A>G, UCN g.1329T>C, CRHR2 c.*13T>C) related to the hypothalamic pituitary adrenocortical (HPA) axis, one of the main stress-response systems, with various stress- and aggression-related parameters at slaughter. These parameters were: physiological measures of the stress response (plasma concentrations of cortisol, creatine kinase, glucose, and lactate), adrenal weight (which is a parameter reflecting activity of the central branch of the HPA axis over time) and aggressive behaviour (measured by means of lesion scoring) in the context of psychosocial stress of mixing individuals with different aggressive temperament. Results: The SNP NR3C1 c.*2122A>G showed association with cortisol concentration (p = 0.024), adrenal weight (p = 0.003) and aggressive behaviour (front lesion score, p = 0.012; total lesion score p = 0.045). The SNP AVPR1B c.1084A>G showed a highly significant association with aggressive behaviour (middle lesion score, p = 0.007; total lesion score p = 0.003). The SNP UCN g.1329T>C showed association with adrenal weight (p = 0.019) and aggressive behaviour (front lesion score, p = 0.029). The SNP CRH g.233C>T showed a significant association with glucose concentration (p = 0.002), and the polymorphisms POMC c.293_298del and MC2R c.306T>G with adrenal weight (p = 0.027 and p < 0.0001 respectively). Conclusions: The multiple and consistent associations shown by SNP in NR3C1 and AVPR1B provide convincing evidence for genuine effects of their DNA sequence variation on stress responsiveness and aggressive behaviour. Identification of the causal functional molecular polymorphisms would not only provide markers useful for pig breeding but also insight into the molecular bases of the stress response and aggressive behaviour in general.

Broiler physiological response to low phosphorus diets at different stages of production

Summary: Phosphorus inclusion in broiler diets needs to be tailored to their physiological demands at different developmental stages. A timed nutritional conditioning strategy, such as P depletion, may enhance mineral homeostasis mechanisms and reduce dietary P supply. This study investigated the effects of dietary P depletion on broiler chickens at different developmental stages, revealing marked responses early on, but indications of compensatory mechanisms later. Endocrine control mediated by calcitriol, intestinal P uptake, and mineral mobilization from bones were identified as potential mechanisms for maintaining mineral homeostasis. Precise timing, duration, and extent of P depletion should be considered for optimized nutrient utilization in broiler husbandry.

Summary: We have developed a paper-based experimental platform for analyzing dengue virus, which incorporates screen-printing technology and black carbon conductive ink. The platform utilizes both two-electrode and three-electrode systems, with an LOD of 0.1 μg/mL for detecting the virus. The three-electrode system exhibits a significantly higher current range and amplification compared to the two-electrode system, making it a viable option for sensitive detection of dengue virus and other diseases.

Efficacy of dietary vitamin D3 and 25(OH)D3 on reproductive capacities, growth performance, immunity and bone development in pigs

Summary: Vitamin D-3 (Vit D-3) and 25(OH)D-3 are used as dietary sources of active vitamin D in pig husbandry, and while their primary effects are on the intestine, kidney, and bone, they also have wide-ranging effects on peripheral tissues. The use of Vit D-3 and 25(OH)D-3 in pig nutrition has been shown to have different outcomes in terms of reproductive capacities, growth performance, immunity, and bone development.

Summary: Improving the nitrogen-utilization efficiency (NUE) of dairy cows through breeding selection has benefits in terms of nutrition, environment, and economics. The individual milk urea concentration (MU) of cows has been proposed as an indicator trait for NUE. This study aimed to identify different microbial genera in the rumen of Holstein cows with divergent genomic breeding values for MU (GBVMU). The identified microbial genera were further investigated for their correlations with MU and other NUE-associated traits, providing insight into genetically determined N-utilization in Holstein cows.

Metabolic Pathway Modeling in Muscle of Male Marathon Mice (DUhTP) and Controls (DUC)-A Possible Role of Lactate Dehydrogenase in Metabolic Flexibility

Summary: This study investigated the contribution of non-mitochondrial and mitochondrial metabolic pathways in contracting muscles during repeated training. The results showed that marathon mice displayed increased glucose and fatty acid oxidation, reduced lactate dehydrogenase activity, and inhibited lactate formation compared to trained control mice. This suggests that marathon mice have enhanced metabolic flexibility towards lipid catabolism regulated by lactate dehydrogenase.

US Swine Industry Stakeholder Perceptions of Precision Livestock Farming Technology: A Q-Methodology Study

Summary: This study analyzes the perceptions of precision livestock farming (PLF) technology held by stakeholders in the US swine industry using the Q-methodology approach. By following up with stakeholders interviewed 6 months earlier, the study found three distinct viewpoints: PLF improves farm management, animal welfare, and labor conditions; PLF does not solve industry problems; PLF has limitations and could lead to data ownership conflicts. Stakeholders with in-depth knowledge of PLF were optimistic about its potential, while those with basic understanding were skeptical. However, all stakeholders agreed on the importance of training and adoption of PLF. The study's results hold promise for informed decision-making about PLF implementation in the US swine industry.

Transcriptomic signature related to poor welfare of sport horses

Summary: The improvement of horse welfare through housing conditions has raised concerns about the detrimental effects of individual housing on horse health and behavior. In this study, blood transcriptome analysis was performed on sport horses housed individually to identify the molecular pathways associated with behavioral changes and welfare impairment. Aggressiveness towards humans was found to be the most correlated behavioral indicator with blood gene expression, mainly related to systemic inflammation. Stereotypies were also correlated with blood gene expression when combined with fecal microbiota analysis. Further studies are needed to understand the biological correlates of unresponsiveness to the environment and alert postures.

Selection for social genetic effects in purebred pigs improves behaviour and handling of their crossbred progeny

Precision livestock farming: a qualitative exploration of swine industry stakeholders

Summary: Precision livestock farming (PLF) technology development has been rapid, but adoption on farms has been slow. Understanding stakeholders' views, applications, limitations, and concerns regarding PLF technology is crucial for understanding adoption factors. Through online interviews, 12 influential stakeholders' views and experiences were investigated. Two distinct stakeholder groups were identified, the PLF enthusiasts and the PLF cautious. The majority of participants were familiar with and experienced in using PLF technologies in the swine industry. PLF technology was perceived to improve decision-making, care for animals, labor efficiency, and resource-use efficiency. Poor internet connectivity and the need to prioritize daily farm tasks were identified as key obstacles to implementation. Stakeholders' concerns included data privacy and the impact of PLF technology on human-animal relationships and farmers' duty of care. Stakeholders prioritized camera systems for monitoring pig health and welfare and ensuring individual pig identification in future PLF development.

Genetic associations between human-directed behavior and intraspecific social aggression in growing pigs

Summary: This study estimated the genetic parameters of human-directed behavior and intraspecific social aggression in growing pigs and found that these traits are heritable and potentially useful for genetic selection. Additionally, there were genetic associations between aggression and fear/boldness indicators. Selecting against extreme shyness and boldness could result in easier to handle pigs during routine farm procedures.
