Summary: Idiopathic pulmonary fibrosis (IPF) is a highly aggressive and poorly understood lung disease, with lung epithelial cells playing a critical role in its initiation and progression. NUMB protein is necessary for the pathological activation of beta-catenin signaling in lung epithelial cells following bleomycin-induced injury. Depletion of Numb and Numblike reduces fibrotic lesion accumulation, preserves lung function, and increases survival rates in mice treated with bleomycin.

Summary: Light-sheet or selective plane illumination microscopy (SPIM) is suitable for in toto imaging of living specimens at high temporal-spatial resolution, but faces limitations due to light scattering in opaque specimens. The Multiview tiling SPIM (MT-SPIM) combines the strengths of Multi-view SPIM (M-SPIM) and a confined, multi-tiled light sheet, providing high-resolution, robust, and rotation-free imaging of living specimens. It improves axial resolution by a factor of two, enhancing automated segmentation of cellular structures.

Summary: This study aimed to discover and validate a predictive panel for surgical outcome of residual tumor mass after first-line debulking surgery. The Nanostring method and immunohistochemistry testing were used to validate a set of predictive genes and predict the presence of miliary carcinomatosis patterns. The results showed that tissue-based biomarkers cannot reliably predict post-operative residual tumor.

However, there is a lack of accurate validation and standardization across laboratories. This study shows that the choice of reference materials during validation significantly affects the analytical performance evaluation of liquid biopsy NGS assays. The findings provide guidelines for selecting suitable reference materials and ensuring comparable results for patients.
