Antiurolithic activity of Origanum vulgare is mediated through multiple pathways

Background: Origanum vulgare Linn has traditionally been used in the treatment of urolithiasis. Therefore, we investigated the crude extract of Origanum vulgare for possible antiurolithic effect, to rationalize its medicinal use. Methods: The crude aqueous-methanolic extract of Origanum vulgare (Ov.Cr) was studied using the in vitro and in vivo methods. In the in vitro experiments, supersaturated solution of calcium and oxalate, kidney epithelial cell lines (MDCK) and urinary bladder of rabbits were used, whereas, in the in vivo studies, rat model of urolithiasis was used for the study of preventive and curative effect. Results: In the in vitro experiments, Ov.Cr exhibited a concentration-dependent (0.25-4 mg/ml) inhibitory effect on the slope of nucleation and aggregation and also decreased the number of calcium oxalate monohydrate crystals (COM) produced in calcium oxalate metastable solutions. It also showed concentration-dependent antioxidant effect against DPPH free radical and lipid peroxidation induced in rat kidney tissue homogenate. Ov.Cr reduced the cell toxicity using MTT assay and LDH release in renal epithelial cells (MDCK) exposed to oxalate (0.5 mM) and COM (66 mu g/cm(2)) crystals. Ov.Cr relaxed high K+ (80 mM) induced contraction in rabbit urinary bladder strips, and shifted the calcium concentration-response curves (CRCs) towards right with suppression of the maximum response similar to that of verapamil, a standard calcium channel blocker. In male Wistar rats receiving lithogenic treatment comprising of 0.75% ethylene glycol in drinking water given for 3 weeks along with ammonium chloride (NH4Cl) for the first 5 days, Ov.Cr treatment (10-30 mg/kg) prevented as well as reversed toxic changes including loss of body weight, polyurea, crystalluria, oxaluria, raised serum urea and creatinine levels and crystal deposition in kidneys compared to their respective controls. Conclusion: These data indicating the antiurolithic activity in Ov.Cr, possibly mediated through inhibition of CaOx crystallization, antioxidant, renal epithelial cell protective and antispasmodic activities, rationalizes its medicinal use in urolithiasis.

Summary: The study aimed to enhance the oral bioavailability of the poorly water-soluble drug tacrolimus by developing niosomes using polyoxyethylene lauryl ether (Brij-35) and poloxamer 407 surfactants. The niosomes were prepared using a thin film hydration technique and evaluated using various techniques including FTIR, ultra-centrifugation, DLS experiments, SEM microscopy, and DSC technique. The results showed that the newly developed niosomes can improve the dissolution profile of tacrolimus and can be considered as potential oral carriers.

Toxicity profiling of Burgmansia aurea Lagerh. Leaves using acute and sub-acute toxicity studies in rats

Summary: This study aimed to detect any hazardous effects of B. aurea on animals and find out its LD50. The results revealed no mortality and morbidity in both acute oral and sub-acute toxicity studies. However, oxidative stress markers were significantly increased in the treatment groups, and animals showed decreased food and water intake in the acute oral toxicity study. A slight difference in renal function tests was observed, but no significant change in histopathology was found in selected organs. This study concluded that B. aurea can be safely used for pharmacological purposes.

An insight into the hepatoprotective role of Velpatasvir and Sofosbuvir per se and in combination against carbon tetrachloride-induced hepatic fibrosis in rats

Summary: This study aimed to examine the antifibrotic effect of direct-acting antiviral drugs Sofosbuvir and Velpatasvir in a rat model of carbon tetrachloride-induced fibrosis. The results showed that Sofosbuvir and Velpatasvir, alone or in combination, significantly inhibited fibrosis and mediated this effect by reducing the levels of TNF-a/NF-& kappa;B and the IL-6 signaling pathway. Therefore, Sofosbuvir and Velpatasvir may have antifibrotic effects independent of their antiviral activity.

An insight investigation to the antiurolithic activity of Trachyspermum ammi using the in vitro and in vivo experiments

Summary: The crude extract of Trachyspermum ammi seeds (Ta.Cr) has shown antiurolithic activity through diuretic effects, inhibition of calcium oxalate crystal aggregation, antioxidant effects, renal epithelial cell protection, and antispasmodic effects. This suggests that Ta.Cr has therapeutic potential in urolithiasis.

Use of Complementary and Alternative Medicine in the General Public of Western Saudi Arabia: A Cross-Sectional Survey

Summary: This study investigated the awareness, self-use, perceptions, beliefs, and attitudes towards complementary and alternative medicine (CAM) in the general public of Jeddah, Saudi Arabia. The majority of the population was aware of various CAM modalities and considered prayers and spirituality to be the most effective. Information about CAM was mainly obtained from friends and relatives, while lack of knowledge and lack of trained professionals were perceived as barriers to CAM implementation. Gender, educational level, and relationship to the medical field were found to be associated with awareness and self-use of CAM modalities.

Despite the belief that herbal medicines are safe and do not require consultation, there is a need to increase awareness and establish strategies to evaluate their safety and efficacy.
