Regulation of sonic hedgehog-GLI1 downstream target genes PTCH1, Cyclin D2, Plakoglobin, PAX6 and NKX2.2 and their epigenetic status in medulloblastoma and astrocytoma

Background: The Sonic hedgehog (Shh) signaling pathway is critical for cell growth and differentiation. Impairment of this pathway can result in both birth defects and cancer. Despite its importance in cancer development, the Shh pathway has not been thoroughly investigated in tumorigenesis of brain tumors. In this study, we sought to understand the regulatory roles of GLI1, the immediate downstream activator of the Shh signaling pathway on its downstream target genes PTCH1, Cyclin D2, Plakoglobin, NKX2.2 and PAX6 in medulloblastoma and astrocytic tumors. Methods: We silenced GLI1 expression in medulloblastoma and astrocytic cell lines by transfection of siRNA against GLI1. Subsequently, we performed RT-PCR and quantitative real time RT-PCR (qRT-PCR) to assay the expression of downstream target genes PTCH1, Cyclin D2, Plakoglobin, NKX2.2 and PAX6. We also attempted to correlate the pattern of expression of GLI1 and its regulated genes in 14 cell lines and 41 primary medulloblastoma and astrocytoma tumor samples. We also assessed the methylation status of the Cyclin D2 and PTCH1 promoters in these 14 cell lines and 58 primary tumor samples. Results: Silencing expression of GLI1 resulted up-regulation of all target genes in the medulloblastoma cell line, while only PTCH1 was up-regulated in astrocytoma. We also observed methylation of the cyclin D2 promoter in a significant number of astrocytoma cell lines (63%) and primary astrocytoma tumor samples (32%), but not at all in any medulloblastoma samples. PTCH1 promoter methylation was less frequently observed than Cyclin D2 promoter methylation in astrocytomas, and not at all in medulloblastomas. Conclusions: Our results demonstrate different regulatory mechanisms of Shh-GLI1 signaling. These differences vary according to the downstream target gene affected, the origin of the tissue, as well as epigenetic regulation of some of these genes.

Summary: Glioblastoma multiforme (GBM) is the most common and aggressive malignant brain tumor with poor prognosis. HDAC6 is overexpressed in GBM and its silencing inhibits proliferation, migration, and clonogenic potential in GBM cell lines, as well as reversing the mesenchymal phenotype, decreasing autophagy, inhibiting the Sonic Hedgehog pathway, and restoring primary cilia expression. These findings suggest HDAC6 may be a promising therapeutic target for GBM treatment.

Regulating gene expression and erythropoiesis, as well as adjusting iron metabolism, can be potential targets for managing thalassaemia.

Differential role of Pax6 and its interaction with Shh-Gli1-IDH2 axis in regulation of glioma growth and chemoresistance

Summary: This study explored the potential interaction and regulation of Sonic hedgehog (Shh)-Gli1 cell signaling with Pax6 and IDH2 in glioma. It was found that Gli1 had differential dynamic interactions with IDH2 and Pax6, and Gli1 knockdown resulted in downregulation of Pax6 and upregulation of IDH2 expression. Additionally, the knockdown of Pax6 gene in glioma cells downregulated the expression of Gli1 and IDH2 and promoted cell proliferation. The study also showed that the combination of Gli1 inhibitor and an anti-cancer drug had a higher efficacy in treating glioma cells.

Summary: This study reports the discovery of a highly potent antibody, P4A2, that can neutralize all circulating variants of SARS-CoV-2, including Omicron. It demonstrates broad neutralizing efficacy and prophylactic and therapeutic effectiveness in mice models. Due to its overlap with the ACE2 binding site, P4A2 may also be highly effective against future variants.

A Comprehensive Review of miRNAs and Their Epigenetic Effects in Glioblastoma

Summary: Glioblastoma, a highly aggressive brain tumor originating from glial cells, has a maximum life expectancy of 14.6 months. Despite multiple promising therapies, clinical outcomes for glioblastoma patients are poor due to drug resistance. Epigenetic modification, particularly DNA methylation, and miRNAs have been identified as major factors in glioblastoma progression and drug resistance. This review aims to explore the role of known miRNAs in glioblastoma, how epigenetic modification affects their expression, and their potential as predictors of treatment response.

Genetic association analysis of rs662799 (-1131A > G) polymorphism of APOA5 gene with morphometric and physio-metric traits using multiplex PCR

Summary: This study investigated the association of genetic variant rs662799 of the APOA5 gene with body mass index (BMI) and blood pressure (BP). The results showed a significant association between this variant and increased BMI and BP, specifically in individuals with the G allele. This study provides important clinical and research implications for the diagnostic and prognostic value of the gene in question.

Analysis of transcriptomic data sets supports the role of IL-6 in NETosis and immunothrombosis in severe COVID-19

Summary: The study found that the expression of NETosis functional genes in the blood of severe COVID-19 patients increased with disease severity, and showed a negative correlation with blood oxygen saturation. This was also validated in the lungs of non-survivors. Additionally, the early increase of IL-6 in severe COVID-19 disease was found to be associated with complement activation.
