Low NKp30, NKp46 and NKG2D expression and reduced cytotoxic activity on NK cells in cervical cancer and precursor lesions

Background: Persistent high risk HPV infection can lead to cervical cancer, the second most common malignant tumor in women worldwide. NK cells play a crucial role against tumors and virus-infected cells through a fine balance between activating and inhibitory receptors. Expression of triggering receptors NKp30, NKp44, NKp46 and NKG2D on NK cells correlates with cytolytic activity against tumor cells, but these receptors have not been studied in cervical cancer and precursor lesions. The aim of the present work was to study NKp30, NKp46, NKG2D, NKp80 and 2B4 expression in NK cells from patients with cervical cancer and precursor lesions, in the context of HPV infection. Methods: NKp30, NKp46, NKG2D, NKp80 and 2B4 expression was analyzed by flow cytometry on NK cells from 59 patients with cervical cancer and squamous intraepithelial lesions. NK cell cytotoxicity was evaluated in a 4 hour CFSE/7-AAD flow cytometry assay. HPV types were identified by PCR assays. Results: We report here for the first time that NK cell-activating receptors NKp30 and NKp46 are significantly down-regulated in cervical cancer and high grade squamous intraepithelial lesion (HGSIL) patients. NCRs down-regulation correlated with low cytolytic activity, HPV-16 infection and clinical stage. NKG2D was also down-regulated in cervical cancer patients. Conclusion: Our results suggest that NKp30, NKp46 and NKG2D down-regulation represent an evasion mechanism associated to low NK cell activity, HPV-16 infection and cervical cancer progression.

Summary: This study analyzed the impact of serum adipocytokines, insulin, and HOMA IR levels on the risk of benign breast disease and breast cancer. The results showed that higher adipsin and visfatin levels in lean women increased the risk of BBD. Additionally, overweight-obese women with higher levels of insulin and HOMA IR had an increased risk of BBD and breast cancer. This suggests that the interaction between adipose tissue and breast epithelium may trigger malignant transformation in overweight-obese women.

Summary: Prostate cancer is the most common tumor in males and the second leading cause of cancer-related deaths. Current screening methods are not sensitive or specific enough, leading to unnecessary interventions. The current prognostic strategies are invasive and there is a lack of tools for predicting tumor progression and treatment response. miRNAs show promise as biomarkers for screening, surveillance, prognosis and treatment response in prostate cancer.

The results showed that IL-21 rs2255979 and IL-21R rs3093301 were associated with a higher risk of RA and could be genetic markers. Additionally, the elevated IL-21 levels in RA patients suggest that IL-21/IL-21R could be a therapeutic target for RA.

Summary: Breast cancer has become the most diagnosed cancer, with a high mortality rate among women. Selective estrogen-receptor modulators (SERMs) have been used for the prevention and treatment of estrogen receptor-positive breast cancer, but their long-term oral administration has been associated with toxicity and side effects. This study explores the use of a polymeric gel-like solution as a topical administration route for the active metabolite of tamoxifen (EDX), which has shown significant antitumor efficacy and reduced toxicity compared to oral tamoxifen. This novel administration route could have a strong impact on breast cancer prevention and treatment.

Summary: The phase angle, an indicator of muscle mass status and membrane cell integrity, has been found to be associated with negative outcomes in women with uterine cervical cancer. In a study with 65 participants, the low phase angle group exhibited higher levels of inflammation and poorer quality of life compared to the normal phase angle group. Additionally, the interaction between phase angle and obesity was found to be an important factor in different histologic subtypes of uterine cervical cancer.

Summary: The molecular profile of breast cancer in Latin-American women was studied in five countries. The study described the socioeconomic characteristics, risk factors, prognostic factors, and molecular subtypes, and estimated the 60-month overall cumulative survival probabilities. Factors such as country of residence, intrinsic subtype, age, and tumor stage were found to be associated with overall survival.
