Summary: This study focused on the development and characterization of short cationic amphiphilic beta-sheet forming anticancer peptides. The peptides exhibited selective anticancer activity influenced by their surface activity and secondary structure in hydrophobic surfaces, leading to disruption of mitochondrial membranes and apoptosis. Insights gained from the study can be used to design new ACPs with improved anticancer activity and lower toxicity against normal cells.

Summary: Cell surface proteoglycans, such as syndecans, play crucial roles in regulating cell behavior, including interactions with extracellular matrix components and mediating proliferation, adhesion, and migration. In tumors, changes in the levels and distribution of syndecan-1 can have implications for prognosis, with loss of membrane staining and abnormal cytoplasmic or nuclear staining often indicating poor outcomes. Targeting syndecan-1 with antibody-toxin conjugates may hold promise for clinical applications in both myeloma and some carcinomas.

Summary: This study aimed to investigate the interplay between heparan sulfate (HS) degradation and radiation response in triple-negative breast cancer (TNBC) cells. The results showed significantly increased radioresistance and cell migration after HS degradation. Furthermore, the expression and activation of certain signaling pathway molecules were altered after HS degradation, impacting the radiation resistance of TNBC cells.

Summary: The study revealed that the presence of heparan sulfate is crucial for the binding between the spike protein of SARS-CoV-2 and host ACE2, while the stability of the T470-F490 loop and hydrophobic interactions play key roles in the binding process.

Summary: In this study, the role of host mucins and mucin glycans on SARS-CoV-2 entry into airway epithelial cells was investigated. The removal of mucins from the surface of cells increased the binding of the virus and enhanced infection. This study demonstrates the important role of glycosylated extracellular mucin domains in the entry of SARS-CoV-2.

Summary: This study investigated the role of HS analogues in modulating exosome secretion, composition, and functions in tumor cells. The findings showed that heparin-induced exosomes significantly inhibited tumor proliferation, migration, and invasion compared to exosomes induced by other HS analogues. Furthermore, the sulfate group at 6-O position of heparan sulfate was identified as playing a crucial role in tumor-derived exosome formation and functions.

Summary: Despite advances in therapeutic approaches, there are still unmet medical needs for patients with multiple myeloma (MM), requiring new treatment strategies. In this study, a phage display technique was used to discover single chain variable fragments (scFvs) that recognize a specific glycoform of an MM surface antigen. These scFvs activated NK cells and induced specific killing of MM cells, suggesting their potential as therapeutic agents. The development of antibodies and peptides that detect tumor-specific glycoforms holds promise for improving targeted therapies and tumor imaging.

Summary: This study found that influenza infection dynamically induces methicillin-resistant Staphylococcus aureus (MRSA) to increase cytotoxin expression while decreasing metabolic pathways. The activity of a specific cytotoxin is shaped by the post-influenza lung microenvironment, as it is activated by epithelial glycocalyx fragments released after influenza infection. The severity of post-influenza MRSA pneumonia is influenced by bidirectional host-pathogen interactions.

Summary: This article describes a method for synthesizing a library of HS mimetics from natural aminoglycosides, reducing the number of steps compared to traditional methods. Computational modeling was used to identify a new class of trisaccharide compounds derived from tobramycin that mimic natural HS and have a strong binding to heparanase but a low affinity for off-target platelet factor-4 protein.
