Suppression of CDK2 expression by siRNA induces cell cycle arrest and cell proliferation inhibition in human cancer cells

Cyclin-dependent kinase 2 (CDK2) is a member of serine/threonine protein kinases, which initiates the principal transitions of the eukaryotic cell cycle and is a promising target for cancer therapy. The present study was designed to inhibit cdk2 gene expression to induce cell cycle arrest and cell proliferation suppression. Here, we constructed a series of RNA interference (RNAi) plasmids which can successfully express small interference RNA (siRNA) in the transfected human cells. The results showed that the RNAi plasmids containing the coding sequences for siRNAs down-regulated the cdk2 gene expression in human cancer cells at the mRNA and the protein levels. Furthermore, we found that the cell cycle was arrested at G0G1 phases and the cell proliferation was inhibited by different siRNAs. These results demonstrate that suppression of CDK2 activity by RNAi may be an effective strategy for gene therapy in human cancers. [BMB reports 2010; 43(4): 291-296]

Tree-ring cellulose oxygen isotopes indicate atmospheric aridity in the western Kunlun Mountains

Summary: Vapor pressure deficit (VPD) is an important indicator of atmospheric aridity, but there is a lack of data to represent VPD globally. In this study, tree-ring cellulose oxygen isotope (δ(OTRC)-O-18) in the western Kunlun Mountains is shown to be a hidden frequency-dependent indication of atmospheric aridity. The results reveal that δ(OTRC)-O-18 values are strongly correlated with VPD at timescales < 5 years.

Cyclin C: A new responser for chemosensitivity in cancer

Tropical and Polar Oceanic Influences on the Cold Extremes in East Asia: Implications of the Cold Surges in 2020/2021 Winter

Summary: The study investigates the winter cooling in East Asia and its associated atmospheric and oceanic influences. The results show that the cooling is characterized by anticyclonic circulation, mid-to-high-latitude wave trains, decreased sea surface temperature in the tropical Pacific, and reduced sea-ice cover in the Barents and Kara Seas. Two circulation modes, influenced by La Nina-type SST anomaly and reduced SIC in BKS, have a cooling effect on East Asia. The synergy between tropical and polar oceanic forcing can increase the East-Asia cooling days and cold extremes.

Triptolide improves chondrocyte proliferation and secretion via down-regulation of miR-221 in synovial cell exosomes

6-Shogaol inhibits the proliferation, apoptosis, and migration of rheumatoid arthritis fibroblast-like synoviocytes via the PI3K/AKT/NF-κB pathway

Summary: This study found that 6-shogaol inhibits proliferation, migration, and invasion, and induces apoptosis in rheumatoid arthritis fibroblast-like synoviocytes (FLSs). It also reduces the production of inflammatory cytokines and matrix metalloproteinases (MMPs). Molecular mechanism analysis revealed that 6-shogaol inhibits the PI3K/AKT/NF-kappa B pathway by activating PPAR-gamma. Treatment with 6-shogaol ameliorated joint destruction in a mouse model of collagen-induced arthritis (CIA). Therefore, 6-shogaol may be a potential natural drug for future rheumatoid arthritis treatments.

Effects of balance training in addition to auxiliary activity on balance function of patients with stroke at high risk for falls

Extreme Cold Events in North America and Eurasia in November-December 2022: A Potential Vorticity Gradient Perspective

Summary: From 17 November to 27 December 2022, frequent extremely cold snowstorms occurred in North America and Eurasia. These events were closely related to the establishment of blocking circulations. The Alaska Blocking (AB) and subsequent Ural Blocking (UB) episodes were associated with the phase transition of the North Atlantic Oscillation (NAO) and represented the main atmospheric regimes in the Northern Hemisphere. The dispersion and propagation of Rossby wave packets from Alaska to East Asia created a large-scale connection between AB and UB episodes.

Emerging role of long non-coding RNA JPX in malignant processes and potential applications in cancers

Summary: Long non-coding RNAs (lncRNAs) are crucial regulators of gene regulation and malignant processes in human cancers. The lncRNA JPX acts as a novel molecular switch for X chromosome inactivation and its differential expression has clinical correlations in various cancers. JPX is involved in cancer growth, metastasis, and chemoresistance through its role as a competing endogenous RNA, protein interaction, and regulation of specific signaling pathways. Furthermore, JPX has potential applications as a biomarker and therapeutic target in cancer diagnosis, prognosis, and treatment. This article provides a comprehensive summary of the structure, expression, and function of JPX in malignant cancer processes, and discusses its molecular mechanisms and potential applications in cancer biology and medicine.

Cobalt nanoparticles induce mitochondrial damage and ?-amyloid toxicity via the generation of reactive oxygen species

Summary: Exposure to cobalt nanoparticles (CoNPs) is associated with neurodegenerative disorders, and the mitochondrial mechanisms underlying their neurotoxicity have not been fully characterized. This study found that CoNPs exposure reduced survival and lifespan in nematodes. Additionally, CoNPs exposure aggravated paralysis and beta-amyloid aggregation, accompanied by reactive oxygen species overproduction, ATP reduction, and mitochondrial fragmentation.

Peripheral Blood-derived CircVPS35L as a Potential Diagnostic Biomarker for Non-small Cell Lung Cancer

Summary: Circular RNAs (circRNAs) are dysregulated in cancers and have stable expression in body fluids. This study identified and evaluated the clinical value of circRNA VPS35L (circVPS35L) as a biomarker for diagnosing non-small cell lung cancer (NSCLC). The expression levels of circVPS35L were determined in tissues, blood, and cell lines using RT-qPCR. The stability of circVPS35L was examined with the actinomycin D assay and RNase R treatment. ROC analysis showed that circVPS35L had higher diagnostic value than conventional tumor markers in NSCLC patients. Furthermore, circVPS35L was highly stable in peripheral blood. These findings highlight the potential of circVPS35L as a novel biomarker for NSCLC diagnosis and differentiation from benign lung disease.
