Summary: Immune checkpoint inhibitors have shown success in treating advanced cancers but can also lead to immune-related adverse events (irAE). This study investigated the association between baseline autoantibodies and irAE development in patients receiving ICI therapy for melanoma. Results showed that lower autoantibody concentrations at baseline and a greater change in antibody concentration over time were associated with organ-specific irAE. Further research is needed to understand the role of autoantibodies in the pathogenesis of irAE.

Summary: A retrospective cohort study on patients with pre-existing psoriasis receiving ICIs for cancer treatment showed that most patients experienced psoriasis exacerbation after treatment, but it was manageable with standard therapies; some patients also experienced other immune-related adverse events; patients with disease exacerbation performed well in terms of progression-free survival compared to those who did not.

Summary: For patients with autoantibody-associated psychiatric syndromes, early corticosteroids, intravenous immunoglobulins, or plasmapheresis are recommended as first-line immunotherapy, followed by rituximab or cyclophosphamide as second-line therapy if first-line options fail. Maintenance therapy with mycofenolate mofetil or azathioprine is suggested for those responding to steroids. Retrospective cohort studies support the use of first- and second-line, and maintenance immunotherapies, but more research is needed to establish efficacy and safety through clinical trials.

Summary: The study showed that tumor-associated immune and genomic biomarkers of response to combined immune checkpoint blockade (CICB) in melanoma patients were similar to those of monotherapy, while toxicity from CICB was associated with a more diverse peripheral T-cell repertoire. Additionally, analysis of gut microbiota revealed a higher abundance of Bacteroides intestinalis in patients experiencing toxicity, along with an upregulation of the inflammatory factor IL-1 beta in colitis samples.
